摘 要:目的探讨复方黄黛片(CRNIT)灭活兔血清对人红白血病(HEL)细胞的促凋亡作用及JAK2基因的影响。方法应用PCR探针法鉴定HEL细胞是否存在JAK2突变,制备CRNIT、羟基脲片、生理盐水3组灭活含药兔血清,用不同浓度的血清处理HEL细胞,采用实时荧光定量PCR法检测JAK2基因的表达,采用Western blotting检测相关蛋白的表达。结果 HEL细胞经检测为纯合JAK2 V617F突变;在一定浓度范围内,CRNIT灭活兔血清对HEL细胞JAK2 mRNA表达有抑制作用,羟基脲及生理盐水灭活兔血清对HEL细胞JAK2 mRNA表达无影响;在一定浓度范围内,CRNIT灭活兔血清对HEL细胞Survivin蛋白表达的抑制作用呈浓度依赖性,羟基脲及生理盐水灭活兔血清对HEL细胞Survivin蛋白表达无影响。结论 CRNIT灭活兔血清抑制HEL细胞JAK2 mRNA表达,其意义及作用机制仍需进一步实验研究;CRNIT灭活兔血清抑制HEL细胞增殖,促进HEL细胞凋亡,与抑制Survivin蛋白表达相关。Objective To explore the effects of compound realgar and natural indigo tablet(CRNIT) contained inactivated rabbit serum on apoptosis and JAK2 gene in human erythroleukemia(HEL). Methods PCR probing method was used to identify whether JAK2 mutation was existed or not in HEL cells. Three groups of inactivated drug-contained rabbit serum were prepared,named CRNIT group,hydroxyurea tablet group and physical saline group. HEL cells were treated with the serum of different concentrations. The real-time fluorescence quantification PCR method was adopted to determine the expression of JAK2 gene. Western blotting method was applied to determine the relevant protein expression. Results The JAK2 V617 F mutation happened in HEL cells. In a certain concentration range,the inactivated CRNIT-contained rabbit serum inhibited JAK2 mRNA expression in HEL cells. The inactivated rabbit serum with hydroxyurea and physical saline contained had no effect on JAK2 mRNA expression in HEL cells. In a certain concentration range,the inactivated CRNIT-contained rabbit serum presented the concentration dependence in the inhibition of survivin protein expression in HEL cells. The inactivated rabbit serum with hydroxyurea and physical saline contained had no effect on survivin protein expression. Conclusion The inactivated CRNIT-contained rabbit serum inhibits JAK2 mRNA expression of HEL cells and the significance and effect mechanism need to be further verified in the experimental research. The inactivated CRNIT-contained rabbit serum inhibits HEL cell proliferation and promotes HEL apoptosis,which is related to the inhibition of survivin protein expression.
