摘  要:目的观察去甲肾上腺素(NA)对大鼠背根神经节(DRG)神经元γ-氨基丁酸(GABA)A受体(GABAAR)的表达和功能的影响。方法将离体培养的大鼠DRG神经元分为三组,分别为浓度组、时间组和对照组。其中浓度组各培养瓶中分别加入不同浓度的NA(0.01μmol/L、0.1μmol/L、1μmol/L、10μmol/L),隔日添加一次,第三天进行蛋白测定;时间组各培养瓶中加入1μmol/L的NA,分别于不同时间(5 h、12 h、24 h、48 h、96 h)进行蛋白测定;对照组不做任何处理。运用Western blot法进行蛋白测定。全细胞膜片钳技术观察GABAAR电流变化。结果浓度组与对照组蛋白测定结果比较P>0.05;时间组与对照组蛋白测定结果比较P>0.05;施加GABAAR特异性激动剂蝇蕈醇(Mus),DRG神经元上记录到呈浓度依赖性的内向电流(92.6%,75/81),该电流可被GABAAR特异拮抗剂荷包牡丹碱(Bic)所阻断;施加预孵育NA,GABAAR电流被可逆性地抑制(79.7%,55/69),与对照组相比,100μmol/L Mus引起的GABAAR电流可被NA抑制至对照组的65.94±2.53%。结论 NA对GABAAR电流有明显的抑制作用,对GABAAR的表达无影响。Objective To observe the effect of NA on the expression and function of GABAAR in rat DRG neurons. Methods Rat DRG neurons cultured in vitro were divided into three groups: concentration group, time group and control group. In the concentration group, different concentrations of NA(0.01, 0.1, 1, 10μmol/L) were added to each flask every other day. The protein was measured on the third day. 1μmol/L of NA was added to each flask in the time group. Protein determinations were performed at the time of 5 h, 12 h, 24 h, 48 h and 96 h. No treatment was performed in the control group. Western blot was used for protein determination. Whole cell patch clamp technique was used to observe GABAAR current changes. Results There was no significant difference in protein concentration between the concentration group and the control group, P〉0.05; there was also no significant difference in protein concentration between the time group and the control group, P〉0.05. GABAAR-specific agonist Mus was applied. A concentration-dependent inward current(92.6%, 75/81) was recorded on DRG neurons, which was blocked by the GABAAR-specific antagonist Bic. After preincubation of NA, GABAA current was reversibly inhibited(79.7%, 55/69), compared with the control group, GABAA current induced by 100μmol/Lmol/L Mus was inhibited by NA to 65.94±2.53% of the control group. Conclusion NA can significantly inhibit the GABAAR current and have no effect on the expression of GABAAR.
