摘  要:目的:研究当归的有效成分阿魏酸、黄芪有效成分黄芪甲苷及其配伍对大鼠肝星状细胞HSC-T6脂质过氧化物(LPO)含量、抗氧化酶及Ⅱ相解毒酶的影响。方法:阿魏酸、黄芪甲苷单独及联合处理HSC-T6细胞24 h,分光光度法检测HSC-T6细胞LPO含量和超氧化物歧化酶((SOD)活性,Western Blot检测HSC-T6细胞谷氨酰半胱氨酸合成酶催化亚基(GCLc)、谷氨酰半胱氨酸合成酶调节亚基(GCLm)、血红素加氧酶1(HO-1)、SOD和磷酸化糖原合成酶激酶3β(p-GSK-3β)蛋白表达,实时定量PCR检测HSC-T6细胞GCLc、GCLm、HO-1和SOD1 mRNA表达。结果:黄芪甲苷显著抑制了HSC-T6细胞中LPO含量,增加了SOD活性,上调了GCLc和SOD的mRNA和蛋白表达,增加了GSK-3β的磷酸化水平(均P〈0.05),而对GCLm表达无显著性影响(P〉0.05)。阿魏酸对这些指标均无显著性影响(均P〉0.05)。与黄芪甲苷处理组比较,联合配伍的作用效果无显著性提高(均P〉0.05)。结论:当归补血汤的有效组分黄芪甲苷通过上调Ⅱ相酶的表达抑制了肝星状细胞氧化应激。Objective: This study aims at evaluating the effect of active component compatibility of Danggui Buxue Decoction on oxidative stress and"phase-II"enzymes in hepatic stellate cell( HSC)-T6. Methods: HSC-T6 cells were treated with Astragaloside Ⅳ and/or Ferulic acid for 24 h,and then cells were collected. Lipid peroxidation( LPO) level and superoxide dismutase( SOD) activity were determined using the spectrophotometric method. Protein expression of glutamate-cysteine ligase catalytic subunit( GCLC),glutamate cysteine ligase modifier subunit( GCLm),heme oxygenase-1( HO-1),SOD,and p-glycogen synthase kinase 3β( GSK-3β) were analyzed by Western Blot. Messenger RNA expressions of GCLc,GCLm,HO-1 and SOD1 were analyzed by Real-time polymerase chain reaction( PCR).Results: Astragaloside Ⅳ but not Ferulic acid inhibited LPO levels,elevated SOD activity,up-regulated the protein and mRNA expressions of GCLc and SOD1,increased GSK-3β phosphorylation,while Astragaloside Ⅳ did not significantly modify GCLm expression in HSC-T6 cells. The combination treatment of Ferulic acid and Astragaloside Ⅳ failed to cause a greater effect than Astragaloside Ⅳ alone. Conclusions: Astragaloside Ⅳ,an active component of Danggui Buxue Decoction,inhibits oxidative stress through up-regulation of "phase-II"enzymes expression in HSC-T6.
