摘 要:目的:通过观察香砂六君子丸对脾虚痰浊大鼠肝脏JAK2/STAT3/Bcl-2途径的影响,探讨健脾化痰法防控肥胖人群罹患肝病风险的可能性。方法:40只雄性SD大鼠,随机分为对照组、脾虚痰浊组(模型组)、香砂六君丸低剂量治疗组(香低组)、香砂六君丸中剂量治疗组(香中组)和香砂六君丸高剂量治疗组(香高组),每组8只;ELISA法检测肝组织谷丙转氨酶(GPT)和谷草转氨酶(GOP)水平、血清超氧化物岐化酶(SOD)、丙二醛(MDA)和脂多糖(LPS)浓度以及谷胱甘肽过氧化物酶(GSH-Px)活性;免疫组化法检测肝组织磷酸化(p)-JAK2、p-STAT3和Bcl-2表达。结果:与对照组比较,模型组GPT、GOT、MDA和LPS水平上升,SOD水平和GSH-Px活性下降,p-JAK2/STAT3表达上升,Bcl-2表达下降,均有统计学意义;与模型组比较,后2组GPT、GOT和MDA以及后3组的LPS水平下降,但后3组的SOD水平和GSH-Px活性上升,香高组的p-JAK2和后2组的p-STAT3表达下降,香高组的Bcl-2表达增加,均有统计学差异。结论:健脾化痰法可以对抗氧化应激和内毒素血症,干预JAK2/STAT3/Bcl-2途径,可能具有控制肥胖人群罹患肝病的作用。Objective: To study effects of Xiangsha Liujun Pills( XLP) on expressions of JAK2,STAT3 and Bcl-2 in obesity rats liver tissue,attempting to explore the possibility of the principle of invigorating spleen and resolving phlegm controlling the development liver diseases in obesity. Methods: Forty male SD rats were randomly divided into the control group,spleen deficiency and phlegm-turbid group( model group),XLP low-dose treating group( XLG),XLP middle-dose treating group( XMG) and XLP high-dose treating group( XHG),8 rats in each group. ELISA was used to measure glutamic-pyruvic transaminase( GPT) and glutamic-oxalacetic transaminease( GOP) levels in liver tissue,and serum levels of superoxide dismutase( SOD),malondialdehyde( MDA) and lipopolysaccharide( LPS) and glutathion peroxidase( GSH-Px) activity. The immunohistochemistry was used to observe phosphorylated( p)-JAK2,p-STAT3 and Bcl-2 expressions in liver tissue. Results: Compared with those in the control,SOD level,GSH-Px activity and Bcl-2 expression were decreased,but levels of MDA,LPS,GTP and GOP,and p-JAK2/STAT3 expressions were increased significantly in the model group. Compared with those in the model group,SOD levels and GSH-Px activity were increased,but LPS level was decreased significantly in later 3 groups. MDA level and p-STAT3 expression in later2 groups were markedly reduced. GOT level in XLG was obviously decreased. The p-JAK2 and Bcl-2 expressions in XHG were significantly reduced and increased. Conclusion: The principle of invigorating spleen and resolving phlegm might control the development of liver diseases in obesity people via resisting oxidative stress and endotoxima and interpreting JAK2/STAT3/Bcl-2 pathway.
