摘 要:目的:研究麻黄水煎液及拆分组分的利尿作用,并初步探讨其作用机制。方法:将水负荷大鼠分为空白对照组,氢氯噻嗪组(0.017 g·kg-1),麻黄水煎液组(2.34 g·kg-1),生物碱组(0.020 g·kg-1),非生物碱组(0.105 g·kg-1),醇沉组(0.132 g·kg-1),挥发油组(0.9333×10-3m L·kg-1),各组给予相应药物,空白对照组给予同体积生理盐水。采用大鼠代谢笼法,观察麻黄水煎液及拆分组分对大鼠尿量的影响;微板法检测尿氯(Cl-)、尿钾(K+)水平;比浊法检测尿钠(Na+)水平;离子选择电极法检测尿渗透压;酶联免疫吸附测定法(ELISA)检测血浆抗利尿激素(ADH),肾脏水通道蛋白2(AQP2)水平;蛋白免疫印迹法(Western Blot)检测肾脏水通道蛋白2(AQP2)水平。结果:与空白对照组相比,麻黄水煎液和生物碱组分能显著增加大鼠尿量和尿液电解质排泄,降低尿渗透压,ADH,AQP2水平(P〈0.01或P〈0.05)。结论:麻黄具有显著的利尿作用,其利尿作用的组分主要为生物碱,且发挥利尿作用的机制可能与降低ADH的水平和AQP2的表达有关。Objective: To study the diuretic effect of Ephedra Decoction and its splitting fractions and explore its mechanism. Methods: The rats were divided into blank control group,hydrochlorothiazide group( 0. 017 g·kg-1),Ephedra Decoction group( 2. 34 g·kg-1) ephedra alkaloid group( 0. 020 g·kg-1),non-alkaloid group( 0. 105 g·kg-1),alcoholic acid group( 0. 132 g·kg-1),volatile oil group( 0. 933 3 × 10-3 m L·kg-1). Each group was given the corresponding drug. The blank control group was given the same volume of saline. The rat metabolites were used to observe the effects of ephedrine decoction and the splitting fractions on the urine volume of rats. The levels of urinary chlorine( Cl-)and urinary potassium( K+) were measured by microplate method. The level of urinary sodium( Na+) was measured by turbidimetric method. The enzyme-linked immunosorbent assay( ELISA) was used to detect plasma antidiuretic hormone( ADH) and kidney channel protein 2 levels. Western Blot was used to detect the level of renal AQP2. Results:Compared with the blank group,Ephedrine Decoction and alkaloid components could significantly increase urine output and urinary electrolyte excretion,decrease urinary osmotic pressure,ADH and AQP2 levels( P〈 0. 01 or P〈 0. 05).Conclusion: Ephedra Decoction has significant diuretic effect and alkaloids are the diuretic part and the mechanism of diuretic action may be related to the reduction of ADH levels and the expression of AQP2.
