黄芪对细胞毒素相关蛋白A诱导异常增殖的大鼠系膜细胞的影响

摘　　要：目的观察黄芪对细胞毒素相关蛋白A(CagA)诱导异常增殖的大鼠肾小球系膜细胞的影响。方法将体外培养的大鼠系膜细胞随机分为空白对照组、IL-1β(10ng/ml)阳性对照组、CagA(4μg/ml)实验组、CagA(4μg/ml)+黄芪(1mg/ml)干预组,给予相应的处理后培养72h,CCK-8法检测4组细胞增殖能力,免疫组织化学法检测增殖细胞核抗原(PCNA)表达水平。结果 IL-1β阳性对照组、CagA实验组细胞增殖能力均较空白对照组明显增强(均P<0.05),PCNA表达水平均较空白对照组明显升高(均P<0.05);CagA+黄芪干预组细胞增殖能力较CagA实验组减弱(P<0.05),PCNA表达水平较CagA实验组降低(P<0.05)。结论黄芪或通过降低PCNA的表达水平,抑制CagA诱导异常增殖大鼠系膜细胞的增殖能力,可为Ig A肾病提供新的治疗思路。Objective To investigate the effects of Astragalus membranaceus （AM） on CagA-induced abnormal proliferationof rat mesangial cells. Methods The cultured rat mesangial cells were divided into4 groups： blank control group; IL-1β（10ng/ml） group; cagA （4滋g/ml） group and AM （1mg/ml） treated group. After 72h the cell proliferation was examined by CCK-8and the expression of PCNA was detected by immunohistochemistry. Results The cell proliferation and the expression of PCNAin IL-1 and CagA groups were significantly increased compared with control group（P〈0.05）; while the cell proliferation andexpression of PCNA in AM treated group were significantly decreased compared with CagA group（P〈0.05）. Conclusion Astra-galus Membranaceus can ameliorates CagA-induced abnormal proliferation of rat mesangial cells by inhibiting PCNA expression,which suggest that AM might be effective for treatment of IgA nephropathy.
