摘 要:目的探讨小柴胡汤对实验性肝损伤大鼠的保护作用以及量效关系。方法将130只Wistar大鼠随机分为7组,其中四组给予不同剂量的小柴胡汤,其余各组分别作为健康对照组、伪实验组、多烯磷脂酰胆碱治疗组。对照组(A组)大鼠不做任何干预,给予正常饲养观察;伪实验组(B组)腹腔注射生理盐水2 ml/kg,每3天1次,共6周,同时给予1.5 ml纯净水灌胃,每天1次;模型组(C组)大鼠及其余各干预组(D组、E组、F组)大鼠给予腹腔注射40%四氯化碳植物油混合溶液,每3天1次,同时,给予D组、E组、F组三组大鼠分别8.5 g/kg、26.5 g/kg、44.5 g/kg小柴胡汤灌胃,每天1次;G组四氯化碳腹腔注射,每3天1次,但是灌胃药物为多烯磷脂酰胆碱溶液143.6 mg/kg。6周时处死大鼠,取肝脏病理切片,分析各组病理损害情况;对Masson染色标本用Image-pro plus6对镜下图片进行图像分析,得到各组胶原纤维面积百分比;检测不同时段大鼠天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总胆红素(TBIL)。结果 A组及B组肝小叶结构完整,肝细胞索排列规则有序;C组示肝小叶结构破坏,广泛脂肪变性及空泡变性;G组肝小叶结构较正常,肝细胞不同程度变性,可见肪肪变性及空泡变性,坏死程度较模型组稍轻;D组、E组、F组病变程度介于C组及G组之间。小柴胡汤治疗组胶原纤维面积百分比低于模型组,差异具有统计学意义(P<0.05);小柴胡汤各治疗组间两两相比,胶原纤维面积百分比均存在统计学差异(P<0.05);小柴胡汤治疗组及多烯磷脂酰胆碱治疗组的ALT、AST、TBIL水平均低于模型组,差异具有统计学意义(P<0.05);大剂量小柴胡汤与多烯磷脂酰胆碱两组作用无显著差异(P>0.05)。结论小柴胡汤对实验性肝损伤大鼠有保护作用,其保护作用存在剂量依赖性,按照经方所述的剂量(大剂量)给药对肝损伤的保护作用最佳,与多烯磷脂酰胆碱作用相当。Objective To investigate the protective effect and dose-effect relationship of Xiao Chaihu Decoction on experimental liver injury rats. Methods 130 Wistar rats were randomly divided into 7 groups, 4 groups were treated with different doses of Xiaochaihu Decoction, the other groups were healthy control group, pseudo experimental group, polyene phosphatidylcholine group. The control group were given no intervention and were given normal feeding. Group B was a pseudo-experimental group, intraperitoneal injection of normal saline 2 ml/kg, once every 3 days for 6 weeks, while 1.5 ml of purified water was administered to the stomach once a day. The model group (group C) and other intervention groups (group D, group E, group F) were given intraperitoneal injection of 40% carbon tetrachloride vegetable oil mixed solution once every 3 days, and group D, group E and group F were given 8.5 g/kg, 26.5 g/kg, 44.5 g/kg Xiaochaihu soup, once a day. Group G was intraperitoneally injected with carbon tetrachloride once every 3 days, but the drug was a polyisophosphatidylcholine solution of 143.6 mg/kg. The rats were sacrificed at 6 weeks, and liver pathological sections were taken to analyze the pathological damage of each group. Image analysis of the images under the microscope with Image-pro plus6 for Masson stained specimens, and the percentage of collagen fibers in each group was obtained. AST, ALT and TBIL of rats in different periods were detected. Results The hepatic lobules in group A and group B were structurally intact, and the hepatic cell cords were arranged in an orderly manner; group C showed hepatic lobular structure destruction, extensive steatosis and vacuolar degeneration. The hepatic lobule structure of group G was normal, and the hepatocytes were degenerated to varying degrees. The degeneration and vacuolar degeneration were observed. The degree of necrosis was slightly lighter than that of the model group. The degree of lesions in group D, group E and group F were between group C and group G. Xiaocha
