摘  要:在pH 5.45的Tris-盐酸介质中,格列齐特与溴化十六烷基吡啶-酸性品红反应生成离子缔合物,致使共振光散射(RLS)显著增强,并产生具有两个较大共振光散射峰的新光谱曲线,这两个共振散射峰分别位于620 nm和370 nm处,在此两波长处,格列齐特在0.006~0.32 mg/L范围内与缔合物的RLS增强强度(△IRLS)呈线性关系,检出限为0.0052 mg/L(单波长法,620 nm)、0.0056(单波长法,370 nm)和0.0027 mg/L(双波长法,620 nm+370 nm),据此建立了测定格列齐特的双波长共振光散射(DWO-RLS)分析法。研究了共振光散射的光谱特征、适宜反应条件、共存物质的影响及实际药品的应用。结果表明,该法用于药物中格列齐特含量的测定,加标回收率和相对标准偏差(RSD)(n=5)分别为99.03%~101.5%和1.1%~1.5%。In a Tris-hydrochloric acid medium of pH 5.45, gliclazide reacts with cetylpyridyl bromideacid magenta to form a ion association, which led to a distinctly enhanced resonance light scattering and the appearance of new resonance light scattering spectral curve with two significantly larger resonant scattering peaks. The two resonant scattering peaks are located at 620 nm and 370 nm, respectively. At the two wavelengths, the mass concentration of gliclazide in the range of 0. 006 to 0.32 mg/L was directly proportional to the resonance light scattering increases intensity ( AIREs ) of system, and the detection limits were 0. 0052 mg/L (single wavelength method, 620 nm) and 0. 0056 mg/L ( single wavelength method, 370 nm) and 0. 0027 mg/L ( dual wavelength method, 620 nm + 370 nm). Based on this, a dual wavelength resonance light scattering (DWO-RLS) method was established for the determination of gliclazide. The spectral characteristics of resonance light scattering, the suitable reaction conditions, the effect of coexisting substances and the application of the actual drugs were also studied. The method had been applied to the determination content of gliclazide in the drugs with spiked recoveries and RSD( n = 5 ) in the ranges of 99. 03 % -101.5% and 1.1% - 1.5 %, respectively.
