摘　　要：为筛选并鉴定与猪脑心肌炎病毒(EMCV) VP1蛋白相互作用的宿主蛋白,本实验采用酵母双杂交系统筛选BHK-21细胞cDNA酵母表达文库。酵母共转化和免疫共沉淀试验表明VP1和RPS20蛋白之间存在特异性相互作用。激光共聚焦试验显示,过表达的VP1和RPS20共定位于细胞质中。本研究初步证实了RPS20与EMCV VP1蛋白之间的相互作用,为进一步研究RPS20蛋白在EMCV感染过程中的生物学功能奠定了基础。VP1 protein is one of the most abundant structural protein of encephalomyocarditis virus（EMCV） particle. It has been demonstrated that VP1 is involved in multiple processes of the EMCV life cycle. In the present study, The ribosomal protein20（RPS20） was identified to be one of the binding partners to VP1 of EMCV screened by yeast two-hybrid（Y2 H） assay using VP1 as the ＂bait＂. The interaction between VP1 and RPS20 was further verified by Y2 H retest and co-immunoprecipitation（Co-IP）experiment. Moreover, confocal microscopy showed that VP1 and RPS20 were colocalized in the cytoplasm in EMCV-infected HeLa cells. Our data showed that RPS20 protein might play a role in the replication processing of EMCV, laying a foundation for further studies of the regulation mechanisms of EMCV life cycle.
