摘 要:目的为了解不同浓度替比夫定对体外培养的人肾小球足细胞(HPC)增殖的影响,初步探讨该药物提高肾小球滤过率的可能机制。方法以HPC为研究对象,设立空白的对照组和不同浓度的实验组(10、20、40、60、100、150 mmol/L),通过倒置显微镜察看细胞形态的变化和4-甲基偶氮唑蓝(MTT)法检测细胞的活力,了解替比夫定对HPC增殖的影响。结果细胞形态的变化:通过倒置显微镜可观察10、60、100 mmol/L浓度组的细胞形状随药物浓度增加及作用时间的延长逐步呈现细胞核的浓缩,甚至呈现胞质空泡;MTT细胞活力检测:当100 mmol/L的药物浓度作用24、48、72 h后细胞抑制率分别为(40.85±1.37)%、(51.03±2.15)%、(76.11±4.40)%,各组间的差异具有统计学意义(P<0.05);替比夫定作用HPC的IC50为100 mmol/L;不同的浓度和不同的作用时间对HPC的抑制率比较差异均有统计学意义(P<0.05)。结论替比夫定对人HPC增殖细胞有明显的抑制作用,呈时间及剂量依赖性。Objective To examine different concentrations of telbivudine on cultured human glomerular podocyte cells( HPC) proliferation,explore the possible mechanism of drugs to improve the glomerular filtration rate. Methods Took HPC as the research object,the establishment of blank control group and experimental groups with different concentrations( 10,20,40,60,100,150 mmol/L),with the inverted microscope to observe cell morphology changes and 4-methyl thiazolyl tetrazolium( methylthiazolyl tetrazolium MTT) method to detect cell viability,to understand the effect of telbivudine on HPC the proliferation. Results The changes of cell morphology with inverted microscope observation can be extended: 10,60,100 mmol/L concentrations in cell shape with the increase of drug concentration and action time gradually showed nuclear enrichment,even showed cytoplasmic vacuoles; MTT cell viability assay: when the drug concentration of 100 mmol/L 24,48,72 h after cell inhibition rate for( 40. 85 ± 1. 37) %,( 51. 03 ± 2. 15) % and( 76. 11 ± 4. 40) %,the difference between groups were statistically significant( P〈 0. 05); telbivudine effect of HPC IC50 100 mmol/L; different concentrations and different time between the inhibitory rate of HPC were different,and the difference had statistical significance( P〈 0. 05). Conclusion Telbivudine on HPC significantly inhibitd the proliferation of cells. The mechanism may be involved in the emergence of renal protective effect in the process of antiviral treatment.
