摘 要:目的探讨槲皮素对鹅膏蕈氨酸(IBO)致痴呆大鼠模型海马神经元的保护作用及机制。方法采用Meynert基底核注射IBO建立痴呆大鼠模型。将90只SD雌性大鼠随机分为对照组,模型组,盐酸多奈哌齐组(阳性药组,3 mg·kg-1),槲皮素高、中、低剂量组(100,30,10 mg·kg-1),连续给药4周。采用Morris水迷宫检测大鼠行为学变化;HE染色观察脑组织病理改变;检测大鼠海马组织中乙酰胆碱(Ach)含量与乙酰胆碱转移酶(Ch AT)活性。结果与对照组比较,模型组大鼠的逃避潜伏期明显延长(P<0.05);大鼠在原平台象限停留时间缩短,站台穿越次数明显减少(P<0.05);大鼠海马区神经元损伤明显;海马组织ACh含量、Ch AT活性均明显降低(P<0.05,P<0.01)。与模型组比较,槲皮素高剂量组的逃避潜伏期明显缩短(P<0.01);槲皮素各给药组对神经元损伤具有一定改善作用,其中槲皮素高剂量组对神经元形态结构改善作用较明显;各给药组的ACh含量、Ch AT活性均有上升趋势,其中槲皮素中剂量组的ACh含量明显升高(P<0.05),槲皮素高剂量组的Ch AT活性显著提升(P<0.01)。结论槲皮素能显著改善IBO致痴呆大鼠模型的认知功能障碍及病理损伤,其作用机制可能与其增加海马区中Ach含量及Ch AT活性有关。Objective To study the protective effect of quercetin on hippocampal neurons in rats with ibotenic acid(IBO)injury and its mechanism.Methods Ninety SD rats were randomly divided into control group,model group,positive drug donepezil(3 mg·kg^-1)group,quercetin(100,30,10 mg·kg^-1)dose groups.Rat model of dementia were established using Meynert basal nucleus injection of ibotenic acid(IBO).Behavioral changes of the rats were observed by Morris water maze;and the brain tissue change was detected by HE staining;the content of acetylcholine(Ach)and acetylcholine transferase(ChAT)were detected using commercial detection kits.Results Compared to the control group,the escape latency of the model group was significantly prolonged(P〈0.05).The duration of rats stayed in the original platform quadrant was shortened,times of platform crossing reduced(P〈0.05),and pathological damage of hippocampus was obvious;Ach content and ChAT activity were significantly lowered(P〈0.05,P〈0.01).Compared with the model group,escape latency of the rats in quercetin high dose group was shortened(P〈0.01).The quercetin treatment groups improved neurons damage to some extent,and the high dose group showed a better improvement.The Ach content and ChAT activity in quercetin treatment groups showed an uptrend and the ACh content was obviously raised in medium dose group(P〈0.05),ChAT activity in high dose group was significantly increased(P〈0.01).Conclusion Quercetin can significantly improve the cognitive function and pathological damage induced by IBO in rats.The mechanism may be related to the increase of Ach content and ChAT activity in hippocampus.
