摘 要:目的建立超高效液相色谱-蒸发光散射检测器法(UPLC-ELSD)测定不同产地知母中新芒果苷、芒果苷、知母皂苷B-Ⅱ、知母皂苷B-Ⅲ、宝藿苷Ⅰ、知母皂苷A-Ⅲ、菝葜皂苷元等7个成分的含量。方法采用UPLC-ELSD法,ACQUITY BEH C18色谱柱(2.1 mm×100 mm,1.7μm);流动相为乙腈(A)-0.2%冰醋酸(B),梯度洗脱;蒸发光散射检测器(ELSD)雾化器(氮气)流量为2.5 L·min^(-1);蒸发温度80℃;载气压力3.14 bar;进样体积10μL。结果 7个化合物分离度良好,呈现较好的线性关系(r>0.999 0),平均加样回收率均在96.98%~102.60%,RSD≤2.80%。结果显示7个被测成分在不同产地之间含量差异较大。结论该方法简单准确,省时高效,可为不同产地知母药材质量控制提供科学依据。Objective To establish a UPLC-ELSD method for simultaneous determination of neomangiferin,mangiferin,timosaponin B-Ⅱ,timosaponin B-Ⅲ,baohuoside-I,timosaponin A-Ⅲand sarsasapogenin in Anemarrhena asphodeloides from different producing areas.Methods UHPLC-ELSD analysis was performed on an UltiMate3000 apparatus with an ACQUITY BEH C18 column(2.1 mm×100 mm,1.7μm),acetonitrile(A)and 0.2%glacial acetic acid(B)were selected as the mobile phase,atomizer(nitrogen)flow rate of evaporative light scattering detector(ELSD)was 2.5 L·min^-1,and the evaporation temperature was 80℃;the gas carrying pressure was 3.14 bar,and the injection volume was 10μL.Results The separation of seven compounds is good with a linear relationship(r〉0.9990).The average sample recoveries were in the range of 96.98%-102.60%with an RSD2.80%.Conclusion The method is rapid,accurate,with high stability and repeatability,which could provide references for the quality control of Anemarrhenae rhizoma.
