摘  要:目的探讨低蛋白摄入对燃煤型氟中毒雌性大鼠卵巢颗粒细胞胰岛素样生长因子-Ⅰ(IGF-Ⅰ)表达的影响。方法将清洁级SD雌性大鼠120只随机分为4组,即对照组、低蛋白摄入组、氟中毒组、氟中毒伴低蛋白摄入组,每组30只。对照组和低蛋白摄入组食用75%非病区玉米饲料(含氟量为5.2 mg/kg),氟中毒组和氟中毒伴低蛋白摄入组食用75%燃煤型氟中毒病区玉米饲料(含氟量96 mg/kg),对照组及氟中毒组饲料中以18 g/kg剂量添加大豆蛋白质,低蛋白摄入组和氟中毒伴低蛋白摄入组按9 g/kg剂量添加大豆蛋白质。动态观察雌性大鼠氟斑牙变化情况,分别于60、120、180 d动情期处死雌性大鼠,以放射免疫法检测雌性大鼠血清雌二醇含量,以免疫组化法检测卵巢IGF-Ⅰ表达。结果成功建立了燃煤型氟中毒伴低蛋白摄入雌性大鼠模型。随着染氟时间的延长,低蛋白氟中毒组和氟中毒组雌性大鼠血清雌二醇水平逐渐降低。染毒60 d时各组雌性大鼠卵巢IGF-Ⅰ表达无明显差异(P>0.05),染毒120、180 d时氟中毒组、氟中毒伴低蛋白摄入组雌性大鼠卵巢IGF-Ⅰ表达均低于对照组,氟中毒伴低蛋白摄入组IGF-Ⅰ表达低于低蛋白摄入组,差异均有统计学意义(P<0.05);染毒120、180 d时雌性大鼠卵巢IGF-Ⅰ含量均低于60 d时,差异有统计学意义(P<0.05)。析因分析结果显示,染毒120、180 d时,单独氟作用对雌性大鼠卵巢IGF-Ⅰ的表达有影响(F值分别为71.85,158.26,P<0.05)。结论燃煤型氟中毒雌性大鼠卵巢颗粒细胞IGF-Ⅰ表达降低,低蛋白摄入对雌性大鼠卵巢IGF-Ⅰ表达无明显影响,两因素对卵巢IGF-Ⅰ的表达无交互作用。Objective To observe the effect of low protein diet on ovary IGF-Ⅰexpression of rats with coal-burning fluorosis.Methods A total of 120 weaned SD healthy rats were divided into four groups randomly: control group, low-protein intake group,fluorosis group, fluorosis+low-protein intake group, 30 rats per group. Control group and low-protein intake group were fed with corn from non-endemic fluorosis area(fluorine content: 5.2 mg/kg), fluorosis group and fluorosis +low-protein intake group were fed with corn from coal-burning fluorosis area(fluorine content: 96 mg/kg). The dose of soybean protein added into feed was 18 g/kg for the control group and fluorosis group, 9 g/kg for the two low-protein intake groups.Dental fluorosis was observed.During the estrus cycle(the 60 th, 120 th, 180 th day of treatment), E2 levels were detected by electrochemical methods and the expression of IGF-Ⅰ in ovarian granulosa cells of rats were detected by immunohistochemistry. Results The animal model of fluorosis was successfully established.With the prolongation of fluoride exposure period, the serum E2 level in the fluorosis +low-protein intake group and the fluorosis group decreased gradually. IGF-Ⅰ expression showed no significant difference among each group at the 60 th day of treatment(P〈0.05), decreased in the fluorosis group and fluorosis+low-protein intake group compared those in the control group at the 120 th day and 180 th day of the treatment(P〈0.05), decreased in fluorosis+low-protein intake group compared with that in low-protein intake group(P〈0.05). The factorial analysis showed that fluoride alone exposure affected the expression of IGF-Ⅰ in ovary significantly at the 120 th day and 180 th day of the treatment(F =71.85, 158.26, P〈0.05). Conclusion The expression of IGF-Ⅰ in ovarian granulosa cells of rats, not affected by low-protein intake,decreases in fluorosis rats, without interaction.
