摘 要:目的观察900 MHz电磁辐射对雄性大鼠睾丸组织氧化损伤和γH2AX蛋白表达的影响。方法将健康6~8周龄清洁级SD雄性大鼠按体重随机分为正常对照组和辐射2、4 h组,每组10只。采用900 MHz电磁辐射,每天1次,连续30 d。测定大鼠睾丸组织MDA、GSH含量及SOD、GSH-Px活力以及γH2AX蛋白的表达。结果与正常对照组比较,各辐射组大鼠睾丸组织MDA含量均增加,而SOD、GSH-Px活力和GSH含量均降低,差异均有统计学意义(P<0.05);且随着辐射时间的延长,大鼠睾丸组织MDA含量呈上升趋势,而SOD、GSH-Px活力和GSH含量均呈下降趋势。无论是免疫组化法还是Western blot法,各辐射组大鼠睾丸组织γH2AX蛋白的表达水平均低于正常对照组,差异均有统计学意义(P<0.05);且随着辐射时间的延长,大鼠睾丸组织γH2AX蛋白的表达水平呈下降趋势。结论 900 MHz电磁辐射可造成大鼠睾丸组织氧化损伤,抑制睾丸组织γH2AX蛋白表达。Objective To explore the effects of 900 MHz electromagnetic radiation on oxidative damage and expression ofγH2 AX protein in testis of male rats. Methods Thirty SD rats were divided into control group,2 h-radiation group,4 h-radiation group(n =10). The control group was given sham exposure, and the radiation group was exposed to 900 MHz electromagnetic radiation(370 μW/cm2) for 0 h,2 h and 4 h per day, concecutively for 30 days respectively. The morphological changes of the testes were observed under light microscope,the levels of MDA,GSH and SOD and GSH-Px in testes were measured by colorimetry,and γH2 AX protein expression was detected by immunohistochemistry and Western blot. Results Compared with the control control group,the content of MDA in the testis of each radiation group increased,but the SOD,GSH-Px activities and GSH content decreased,the differences were statistically significant(P〈0.05); With the prolongation of exposure time, the content of MDA in rat testis showed an upward trend,while the levels of SOD,GSH-Px and GSH showed a downward trend. The expression levels of γH2 AX protein, determined by immunohistochemistry or Western blot, in the testis of rats in each radiation group were lower than those in the control group(P〈0.05); With the prolongation of radiation time,the expression of γH2 AX protein in the testis of rats showed a decreasing trend. Conclusion 900 MHz electromagnetic radiation can cause oxidative damage in the testis of rats,and weaken the expression of γH2 AX protein in the testis.
