摘 要:目的:建立反向高效液相(RP-HPLC)色谱法同时测定延胡索中黄连碱、去氢延胡索甲素、延胡索乙素的含量测定方法。方法:采用Agilent HC-C_(18)(200×4.6mm,5μm)色谱柱,流动相为乙腈-0.3%乙酸水溶液(三乙胺调pH值为5.0),梯度洗脱,流速为1mL·min^(-1),检测波长282nm。结果:40min内延胡索药材中黄连碱、去氢延胡索甲素、延胡索乙素3个成分完全分离,峰面积与浓度呈现良好的线性关系(r=0.9995~0.9999);平均加样回收率在95.5%~98.8%,RSD为2.0%~3.0%。测定延胡索中黄连碱、去氢延胡索甲素、延胡索乙素含量分别为1.94%、2.23%、0.53%。结论:该方法快捷可靠,适用于延胡索中黄连碱、去氢延胡索甲素、延胡索乙素3个生物碱类成分的含量测定。Objective:To establish a reversed phase high performance liquid chromatography (RP-HPLC) method for the coptisine, dehydrofusone and tetrahydropalmatine in Corydalis Rhizomavsimuhaneously.Methods:Agilent HC-C18(200×4.6mm, 5μm) column was used. The mobile phase consisted of acetonitrile-0.3% acetic acid in water (triethylamine adjusted to pH5.0) with gradient elution at a flow rate of 1 mL·min-1, detection wavelength 282nm.Results:In 40 min, the three compounds of coptisine, dehydrofusone and tetrahydropalmatine in Corydalis Rhizoma were completely separated The peak area and concentration showed a good linear relationship (r =0.9995~0.9999). The average recovery rate was 95.5.% ~98.8%, the RSD was 2.0% - 3.0%. The determination of coptisine, dehydrofusone and tetrahydropalmatine in Corydalis Rhizoma were 1.94% ,2.23% ,0.53%respectively.Conclusion:This method is rapid and reliable. It is applicable to the determination the three alkaloids of coptisine, dehydrofusone and tetrahydropalmatine in Corydalis Rhizoma.
