摘 要:光遗传学的发明为神经科学领域带来重大突破,相关技术使对蛋白质功能调控、信号通路激活及动物行为的时空精确控制成为可能,为研究神经回路及信号网络提供了独特的手段。近年来,光遗传学技术与相关报告基因结合被应用到药物筛选平台。该平台利用光信号代替电生理或其他试剂(如生长因子等),激活细胞的受体激酶或离子通道,具有高通量、操作简便及低成本等特点。对光控基因工程技术作为受体酪氨酸激酶的研究手段及新的靶向药物筛选平台进行总结。The invention of optogenetics has brought about a major breakthrough in the field of neuroscience. Related technologies have made it possible to control protein function, signal pathway activation and spatial-temporally control of animal behavior, providing a unique way for studying neural circuits and signal networks. Recently, optogenetics technology and related reporter genes have been applied to drug screening platforms. The platform uses optical signals instead of electrophysiological or other reagents, such as growth factors, to activate receptor kinases or ion channels in cells, with high throughput, ease of operation, and low cost. In this paper, light-control genetic engineering technology as a research tool for receptor tyrosine kinase and a new targeted drug screening platform are summarized.
