生物玻璃复合P-15多肽修复兔颅骨缺损的实验研究

摘  要:目的探讨生物玻璃复合P-15多肽对兔颅骨缺损的修复能力。方法将骨髓基质细胞与复合不同质量浓度P-15多肽(100、200、400μg/mL)的生物玻璃(复合材料)进行体外培养(实验组),并以未复合P-15的生物玻璃作为阴性对照组;接种后1、3、7、10 d检测骨髓基质细胞碱性磷酸酶活性,筛选出多肽质量浓度最适宜的复合材料植入兔颅骨缺损内(复合材料组),植入后4、6、8周通过软X射线和组织学染色观察新骨形成情况。结果 P-15多肽质量浓度为200μg/mL时与生物玻璃共培养的骨髓基质细胞碱性磷酸酶活性明显高于质量浓度为100、400μg/mL的实验组及阴性对照组,差异均有统计学意义(P<0.05);4、6、8周时复合材料组兔颅骨缺损内新骨形成均优于同时间点阴性对照组。结论生物玻璃复合P-15多肽可促进兔颅骨缺损的修复。Objective To investigate the effect of bioglass and peptide /P-15 bone substitute material on the repair of rabbit cranial bone defect. Methods Bioglass was saturated with different concentrations of solution of the P-15 peptide(100,200,400 μg/mL)and cultured with bone marrow stromal cells in vitro(the experimental group). Bioglass without P-15 served as a negative control group. Then the alkaline phosphatase(ALP)activity of bone marrow stromal cells was detected at 1,3,7,10d after inoculation to choose the composite materials with the optimum concentration of P-15 to implant into the cranial bonedefect of rabbit(composite group). The quality and quantity of new bone formation was observe by X-ray and histology stainingat 4th,6th,8th week after implantation. Results At the concentration of 200μg/mL of the P-15 in phosphate buffered saline,the bone marrow stromal cells which were cultured with the Bioglass/P-15 showed greater alkaline phosphatase(ALP)activity than that at the concentrations of 100,400μg/mL in experimental group and the negative control group,and the difference hadstatistical significance(P〈0.05).The new bone formation of cranial bone defect of rabbit at 4th,6th,8th week in the composite group was better than that in the negative group. Conclusion Bioglass/P-15 bone substitute materials can accelerate the repair of rabbit cranial bone defect.
