摘 要:2014年江苏某规模猪场发生腹泻疫情,采集发病仔猪小肠组织及肠内容物,经RT-PCR检测,仅猪流行性腹泻病毒(PEDV)阳性。将病料接种Vero细胞,盲传3代后,出现明显的细胞合胞体病变,细胞空泡化,最终裂解脱落。将细胞反复冻融后,收获病毒,经RT-PCR以及间接免疫荧光(IFA)鉴定后,确认所收获的病毒为PEDV,将该分离毒株命名为CZ2014株(Gen Bank:KT428878.1)。RT-PCR扩增其S基因,序列分析结果表明,CZ2014株S基因序列与近年来流行的毒株同源性较高,而与经典疫苗毒株同源性较低。PEDV流行毒株的分离为该病毒致病机制研究及其防控奠定了基础。In 2014,diarrhea epidemic occurred on a scale pig farm in Jiangsu Province. Samples of the small intestine mucosa and intestinal contents were collected and examined by RT-PCR,and they were PEDV positive. The samples were then inoculated onto Vero cells with10 μg/m L trypsin,and the PEDV CZ2014 strain(Gen Bank: KT428878. 1) was isolated. Phylogenetic analysis was performed,showing that the CZ2014 strain was of low homology to the traditional vaccine strain located in different branches,but more homologous to the epidemic strains isolated in recent years. The present results indicated that the YC2014 strain might be a candidate vaccine strain for PEDV immune control.
