摘  要:【目的】建立青海省海晏县沙化地人工种植菊芋中绿原酸的含量测定方法,并评价青海省海晏县沙化地人工种植菊芋根茎和植物不同部位的绿原酸含量。【方法】采用超高效液相色谱(UPLC)法测定,色谱条件:ACQUITY UPLC BEH C18(2.1mm×100 mm,1.7μm);流动相:乙腈—水梯度洗脱;柱温30℃;检测波长327 nm;流速0.2 m L·min-1;进样量0.5μL。【结果】绿原酸在质量浓度0.006 25~0.1 mg·L-1范围内线性关系良好(R2=0.999 1),平均回收率为98.08%,相对标准偏差(sR)=0.39%。沙化地人工种植1个月后(6月份)时菊芋根中绿原酸平均含量0.13%;人工种植4个月后(9月份)时根中平均含量0.08%,茎、叶平均含量为0.98%。【结论】基于绿原酸含量,结合植株长势和生物产量,青海省海晏县沙化地人工种植菊芋的质量良好;建议综合利用菊芋茎和叶,用于绿原酸提取的原材料之一。Objective To establish a method for determining the content of chlorogenic acid in cultivated Jerusalem artichoke(botanical name being Helianthus tuberosus L.) in sandy land of Haiyan County,Qinghai Province,and to evaluate the content of chlorogenic acid in the rhizome and the other parts of the cultivated Jerusalem artichoke.Methods Ultraperformance liquid chromatography(UPLC)method was used. The chromatographic conditions were as follows:ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7 μm),mobile phase consisting of acetonitrile-water in gradient elution,column temperature at 30 ℃,detection wavelength 327 nm,flow rate 0.2 m L/min,and injection volume 0.5 μL. Results The linear relationship was good in the range of mass concentration of 0.006 25 to 0.1 mg·L^-1(R^2= 0.999 1)of chlorogenic acid,the average recovery was 98.08%,relative standard deviation(sR)was 0.39%. The average content of chlorogenic acid in the Jerusalem artichoke root was 0.13% after cultivation for one month(on June),the average content in roots was 0.08% after cultivation for 4 months(on September),and the average content of stems and leaves was 0.98%. Conclusion Based on the content of chlorogenic acid and combined with plant growth and biological yield,the quality of cultivated Jerusalem artichoke in sandy land of Haiyan County,Qinghai Province is good. It is recommended to comphrehensively use the Jerusalem artichoke stems and leaves as one of the raw materials for chlorogenic acid extraction.
