摘  要:目的探讨白藜芦醇(RSV)对脂多糖(LPS)所致急性肺损伤小鼠内质网应激和肺血管内皮的影响。方法 24只C57BL/6J小鼠随机分为对照(control)组、模型(LPS)组和白藜芦醇(RSV)组;RSV组预先予白藜芦醇(20μg·g^(-1))灌胃7 d,LPS组与RSV组于气管内滴注LPS(5μg·g^(-1))建立肺损伤模型。苏木素-伊红(HE)染色病理检查,酶联免疫吸附(ELISA)法检测肺泡灌洗液(BALF)中白介素(IL)-1β、肿瘤坏死因子(TNF)-α,比色法检测肺组织髓过氧化物酶(MPO)活性,蛋白免疫印迹(Western blot)检测肺组织内质网应激标志蛋白肌醇需求酶-1(IRE-1)、活化转录因子6(ATF6)、葡萄糖调节蛋白78(GRP78)及血管内皮钙黏蛋白(VE-cadherin)、血管细胞黏附分子1 (VCAM1)表达水平。结果 LPS组较control组肺损伤明显,肺组织MPO活性、BALF中炎症因子、白细胞数、总细胞数及蛋白含量增高(P<0.05),肺组织IRE-1α、ATF-6、GRP78表达水平上调(P<0.05),伴随VE-cadherin表达降低及VCAM1表达升高(P<0.05);而RSV灌胃减轻了LPS导致的上述指标变化,具有统计学意义(P<0.05)。结论白藜芦醇可减轻急性肺损伤时肺组织内质网应激和肺血管内皮损伤。Objective The aim of this study was to investigate the effect of resveratrol on lung endoplasmic reticulum stress and pulmonary vascular endothelium in acute lung injury mice. Methods Twenty-four male C57BL/6 mice were randomly divided into control group, LPS group, and RSV group with 8 mice in each group. Mice in RSV group were pretreated with RSV (20 μg·g-1) for 7 days, while acute lung injury was established by lipopolysaccharide (LPS) intratracheal instillation (5 μg·g-1) in LPS group and RSV group. The pathological changes of lung were evaluated by H&E staining. The lung tissue myeloperoxidase (MPO) activities were measured by eolorimetry. The IL-1β level and TNF-α level in BALF were determined by ELISA. The protein levels of IRE-1α, ATF-6, GRP78, VE-cadherin, and VCAM1 in lungs of each group were measured by western blot. Results Severe pathological lung injury was observed in LPS group, as compared with control group. LPS group had increased total and PMN cell counts, total protein levels, IL-1β and TNF-α levels in BALF and lung tissue MPO activities than the control group (P〈0.05), while the IRE-1α ,ATF-6,GRP78 and VCAM1 concentrations were higher and VE-cadhefin concentration was lower in lungs of LPS group than those of control group (P〈0.05). Resveratrol attenuated the damage caused by LPS in RSV group (P〈0.05). Conclusion Resveratrol attenuated injury of pulmonary vascular endothelium in ALI mice and down-regulated the lung endoplasmic reticulum stress.
