摘 要:目的研究七氟烷对大鼠认知功能及海马组织中β-淀粉样蛋白(Aβ)表达的影响。方法将60只SD大鼠随机分为对照组和低、中、高剂量实验组,每组各15只。低、中、高剂量实验组大鼠置于麻醉箱中分别通入1%,2%,4%的七氟烷和氧气的混合气体,对照组仅吸入氧气,均持续吸入6 h。Morris水迷宫系统测定各组老年大鼠认知功能,麻醉处理24 h后用酶联免疫吸附实验(ELISA)检测各组老年大鼠海马组织中Aβ和β-淀粉样物质前体蛋白(APP)蛋白的表达,用蛋白免疫印迹法检测各组大鼠海马组织BACE^(-1)蛋白的表达。结果对照组和低、中、高剂量实验组大鼠第5天逃避潜伏期分别为(10.15±2.63),(18.46±3.15),(18.12±2.85),(34.52±3.69)s。对照组及低、中、高剂量实验组穿台次数分别为(4.61±1.06),(3.02±1.56),(2.81±1.17),(1.28±1.05)次。与对照组比较,低、中、高剂量实验组大鼠第1~5天逃避潜伏期均延长,穿台次数减少(P<0.05);与高剂量实验组比较,低、中剂量实验组大鼠第2~5天逃避潜伏期均缩短,穿台次数增多(P<0.05)。低、中剂量实验组大鼠第1~5天逃避潜伏期及穿台次数差异无统计学意义(P>0.05)。对照组及低、中、高剂量实验组海马组织中Aβ蛋白含量分别为(211.69±9.36),(231.23±10.26),(251.68±43.31),(451.67±40.28)ng·g^(-1),APP蛋白含量分别为(5.26±0.69),(8.28±0.66),(9.26±0.78),(16.27±0.89)ng·g^(-1)。与对照组比较,低、中、高剂量实验组海马组织中Aβ和APP蛋白含量明显升高(P<0.05),且实验组随着七氟烷浓度的升高,海马组织中Aβ和APP蛋白含量逐渐升高(P<0.05)。对照组及低、中、高剂量实验组海马组织BACE^(-1)蛋白相对表达量为0.45±0.09,0.52±0.07,0.67±0.12,1.36±0.25,实验组BACE^(-1)蛋白相对表达量均高于对照组,且随着七氟烷作用浓度的升高海马组织BACE^(-1)蛋白含量逐渐升高(P<0.05)。结论七氟烷对老年大鼠术后认知功能造成一定的影�Objective To investigate the effect of sevoflurane on the cognitive function and the expression of β-amyloid protein( Aβ) in the hippocampus of geriatricrats. Methods Sixty SD rats were randomly divided into control group and low, middle, high dose experimental groups,15 rats in each group. The rats in low,middle,high dose experimental groups were placed the anesthetic box for the mixture of oxygen and 1%,2%,4% sevoflurane, control group only inhaledoxygen. All groups were continuously inhaled for 6 h. Cognitive function of geriatric rats in each group was detected by Morris water maze system. The expression of Aβ and β-amyloid precursor protein( APP) in hippocampus of geriatric rats were detected by enzyme-linked immunosorbent assay( ELISA) at 24 h after anesthesia. Results The escape latency in control group and low, middle, high dose experimental groups on 5 d were( 10. 15 ± 2. 63),( 18. 46 ± 3. 15),( 18. 12 ± 2. 85),( 34. 52 ± 3. 69) s. The numbers of through the platform in control group and low,middle,high dose experimental groups were( 4. 61 ± 1. 06),( 3. 02 ± 1. 56),( 2. 81 ± 1. 17),( 1. 28 ± 1. 05) times.Compared with control group,the 1-5 d escape latency in low,middle,high dose experimental groups were significantly prolonged,the numbers of through the platform decreased( P〈0. 05),compared with high dose experimental group,the 2-5 d escape latency in low,middle dose experimental groups shortened,the numbers of through the platform increased( P〈0. 05),there was no significant difference in the 1-5 d escape latency and the numbers of through the platform between low dose experimental group and middle dose experimental group( P〈0. 05).
