摘 要:目的建立脲酶-谷氨酸脱氢酶偶联法测定游泳池水中尿素的方法。方法游泳池水中的尿素被脲酶催化水解生成的氨与α-酮戊二酸、还原型辅酶I(NADH)和氢离子在谷氨酸脱氢酶的作用下反应生成谷氨酸、NAD+和水,通过在同一条件下测定空白、标准和样品在340 nm处NADH吸光度的降低速率,从而计算出尿素的浓度。结果本法最低检出限为0.2 mg/L,测定范围0.2~10.0 mg/L,灵敏度、精密度、准确度均满足卫生评价标准需要,准确度与国标方法一致,水中铁、锰、铜、锌金属离子、余氯、氨等低于一定浓度时不干扰测定。结论本法灵敏、快速,自动化程度高,各项方法指标符合卫生标准评价的需要,可用于测定游泳池水中尿素。[Objective]To establish urease-glutamic dehydrogenase coupling method for determination of urea in swimming pool water. [Methods] The ammonia produced from the urease hydrolysis reaction of urea in swimming pool water reacted with α-ketoglutarate,NADH and hydrogen ions to produce glutamic acid,NAD+ and water under the action of glutamic dehydrogenase.By measuring the reduction rate of NADH absorbance at 340 nm at the same condition,the concentration of urea was calculated.[Results] The minimum detection limit of this method was 0.2 mg/L,and the range of determination was 0.2-10.0 mg/L. The sensitivity,precision and accuracy all met the needs of the health evaluation standard. The accuracy was in accordance with the national standard method. The determination was not disturbed when the concentrations of iron,manganese,copper,zinc metal ions,residual chlorine and ammonia in water were below a certain concentration. [Conclusion] This method is sensitive,fast and highly automated,and the indicators of all methods meet the needs of health standard evaluation,which can be used for the determination of urea in swimming pool water.
