摘 要:CorA/MRS2/MGT-型镁离子转运蛋白在植物镁离子吸收、转运、分配中有着重要作用。为了探究一个编码蛋白定位于叶绿体的玉米镁离子转运蛋白基因Zm MGT12的功能,对其表达模式、表达量与叶绿素合成间的相关性进行了分析,同时构建了过表达载体并遗传转化了拟南芥。表达分析表明,Zm MGT12在各组织部位中均表达,其中在叶中的表达量最高,大约是根的13倍;此外,Zm MGT12在叶中的表达受光诱导,且具有昼夜节律模式。相关性分析表明,Zm MGT12在叶中的表达量与叶绿素合成具有一定的相关性。转基因分析表明,过表达Zm MGT12没有改变转基因拟南芥植株的生物量、叶绿素含量、叶绿体中镁离子浓度。本研究分析了Zm MGT12详细的表达模式、表达量与叶绿素合成间的相关性,并构建了其过表达载体并对拟南芥进行了遗传转化,有助于确定其在植物中的功能。CorA/MRS2/MGT-type magnesium transporters play vital roles during Mg^2+absorption, transport and allocation in plants. In order to explore the function of maize magnesium transporter Zm MGT12 located in the chloroplast, the correlation between chlorophyll synthesis and its expression patterns and expression levels was analyzed. In addition, the overexpression vector of Zm MGT12 was constructed and genetic transformation was performed in Arabidopsis. Expression analysis showed that Zm MGT12 expressed in different parts but with the highest expression level in leaves, which was about 13 times higher than roots. Moreover, the expression of Zm MGT12 in leaves was induced by light and had circadian pattern. Correlation analysis indicted that there was a slight correlation between the expression level of Zm MGT12 in leaves and chlorophyll synthesis. Transgenic analysis found that the overexpression of Zm MGT12 caused no phenotypic change of transgenic Arabidopsis plants including biomass, chlorophyll content, and the concentration of magnesium in chloroplast. This study analyzed detailed correlation between chlorophyll synthesis and expression patterns and expression levels of Zm MGT12,constructed its overexpression vector and performed genetic transformation of Arabidopsis, which would help to identify its functions in plants.
