青睐苜蓿耐盐突变体愈伤组织过氧化物同工酶特性分析

摘  要:苜蓿是一种高经济价值作物,为了解其耐盐突变体愈伤组织与普通愈伤组织的差异,以突变体与对照为材料,对突变体两个生长阶段的过氧化物同工酶特性进行比较,分析同工酶带型及同工酶各条带过氧化物酶的含量。结果表明:对照苜蓿的两个重复均出现7条酶带p1~p7,耐盐突变体出现5条谱带,缺失的酶带迁移率分别是0.430和0.733。酶谱的相似性系数为0.833。突变体绿色愈伤的主带谱带p1(Rf 0.147)、p2(Rf 0.263)和p7(Rf 0.864)的着色色度和带宽与对照基本相同。说明主要基因一致。各酶带活性分析显示,除p2酶带外,5条共有的酶带中,均为突变体黄色愈伤的酶含量最低。对照苜蓿中迁移率小的p1和p3酶带含量最高,突变体绿色愈伤迁移率大的p4、p7酶含量最高。同工酶总含量中,对照苜蓿最高(56.645 0 mg/m L),突变体绿色愈伤居中(49.653 0 mg/m L),突变体黄色愈伤组织最少(42.806 0 mg/mL)。总之,耐盐苜蓿突变体的过氧化物酶种类和含量的减少说明发生了基因遗传背景上的变异。Alfalfa is a kind of high economic value crop. In order to understand the differences between salt-tolerant mutantcallus (ST mutant) and common variety callus (CK) of alfalfa, the characteristics of peroxidase isoenzymes type and the contentof isozyme were compared between the CK and ST mutant in two development stages of the mutant callus. The results showedthat there were seven zymogram bands(p1-p7) in the CK, while there were only five zymogram bands in ST mutant with lack oftwo bands in Rf 0.430 and Rf 0.733. The similarity coefficient of the zymogram was 0.833. The chromaticity and band width ofthe main isoenzyme bands p1 (Rf 0.147), p2 (Rf 0.263) and p7 (Rf 0.864) of green ST mutant callus were similar with that of CK.It indicated that their major genes were consistent. The analysis of isoenzyme activity showed that the isoenzyme content ofyellow ST mutant callus was the lowest in 5 common isoenzyme bands except for p2 band. The content of p1 and p3 bands withshort mobility was the highest in CK, while the content of p4 and p7 with large mobility was the highest in green ST mutant. Thetotal content of isoenzymes was the highest (56.645 0 mg/mL) in CK, green ST Mutant was in the midium (49.653 0 mg/mL) andyellow ST mutant was the least in (42.806 0 mg/mL). In conclusion, the less isoenzyme bands of ST mutant indicated a variationin the genetic background.
