摘  要:目的观察N-乙酰半胱氨酸(NAC)对哮喘小鼠Th1/Th2失衡及相关转录因子的影响,并讨论其起效机制。方法 6~8周龄SPF级BALB/c小鼠随机分为对照组、模型组、针挑组、NAC低剂量组、NAC中剂量组、NAC高剂量组、阳性组7组,每组15只。模型组、针挑组、NAC低剂量组、NAC中剂量组、NAC高剂量组、阳性组采用卵清蛋白致敏及激发制备哮喘小鼠模型,针挑组在此基础上采用壮医针挑疗法,NAC低剂量组、NAC中剂量组、NAC高剂量组按照10、50、100 mg/kg给予NAC,阳性组给予沙丁胺醇,1次/d,7 d为1个疗程,治疗1个疗程。疗程结束后,进行肺泡灌洗液(bronchoalveolar lavage fluid,BALF)细胞计数; RTPCR法检测T盒转录因子(T-box expressed in T cells,T-bet) mRNA、锌指蛋白3(GATA-binding protein-3,GATA-3) mRNA表达水平; ELISA法检测血清IgE、白细胞介素(IL)-4、IL-5、IL-10、γ干扰素(IFN-γ)、IL-12水平;观察肺组织形态学变化。结果与对照组比较,模型组BALF细胞计数升高,血清Ig E、IL-4、IL-5、IL-10水平升高而IFN-γ和IL-12水平降低,T-bet mRNA表达降低而GATA-3 mRNA表达上升,差异均有统计学意义(P <0. 05)。与模型组比较,针挑组、NAC高剂量组、阳性组BALF细胞计数降低,血清Ig E、IL-4、IL-5、IL-10水平降低而IFN-γ和IL-12水平升高,T-bet mRNA表达上升而GATA-3 mRNA表达降低,差异均有统计学意义(P <0. 05)。结论针挑疗法和高剂量NAC可恢复T-bet和GATA-3表达,进而纠正Th1/Th2失衡,是其治疗哮喘的作用环节之一。Objective To investigate the effects of NAC on Th1/Th2 imbalance and related transcription factors in mice with asthma,and to discuss its therapeutic mechanisms. Methods Six to eight weeks old mice with SPF grade BALB/c were randomly divided into control group,model group,acupuncture group,low-dose NAC group,medium-dose NAC group,high-dose NAC group and positive group( n = 15). The asthma model was established using ovalbumin( OVA) sensitization and stimulating in model group,acupuncture group,NAC groups and positive group. Zhuang acupuncture,NAC treatments and salbutamol were given in acupuncture group,NAC groups and positive group,respectively.After a therapeutic period. The cell count in BALF and the expressions of T-bet mRNA and GATA-3 mRNA were recorded. The levels of serum IgE,IL-4,IL-5,IL-10,IFN-γ and IL-12 were assessed by the method of ELISA.The morphological features of lung tissues were observed. Results Compared with the control group,the cell count in BALF,the levels of serum IgE,IL-4,IL-5 and IL-10,and the expression of GATA-3 mRNA were significantly increased in the model group while the levels of IFN-γ and IL-12,and the expression of T-bet mRNA were significantly reduced( P〈0.01 or P〈0.05). Compared with the model group,the cell count in BALF,the levels of serum Ig E,IL-4,IL-5 and IL-10,and the expression of GATA-3 mRNA were significantly reduced in the acupuncture group and the high-dose NAC group,while the levels of IFN-γ and IL-12,and the expression of T-bet mRNA were significantly increased( P〈0.01 and P〈0.05). Conclusion Acupuncture treatment of Zhuang medicine and the therapy of NAC could regulate the expressions of T-bet mRNA and GATA-3 mRNA,then reverse the imbalance of Th1/Th2 in asthma.
