广佛手对脂多糖诱导的人肺上皮细胞炎症细胞因子调控的影响

摘　　要：目的:探讨广佛手对脂多糖诱导的肺上皮细胞炎症细胞因子的调控作用。方法:不同浓度广佛手水提取物和醇提取物作用于肺上皮细胞A549细胞后,采用CCK8法检测广佛手水提取物和醇提取物对A549细胞活力的影响;以脂多糖(10μg/m L)作用于培养的A549细胞,同时加入广佛手提取物,24 h后ELISA法测定上清液IL-8和IL-10的含量。结果:广佛手水提取物和醇提取物对A549细胞活力影响无统计学意义(P>0. 05);脂多糖作用肺上皮细胞后,IL-8和IL-10释放均增加(P <0. 01),广佛手水提取物和醇提取物可减少脂多糖诱导的IL-8释放增加(P <0. 01),广佛手水提取物在LPS诱导后IL-10分泌明显增加(P <0. 01),而广佛手醇提取物对LPS诱导的IL-10分泌增加呈抑制作用(P <0. 01)。结论:广佛手水提取物和醇提取物对肺上皮细胞无损伤作用,对脂多糖造成的急性肺损伤模型有抑制促炎因子的释放和增加抗炎因子的释放双重保护作用。To investigate the effect of Citrus medica on the regulation of inflammatory cytokines in pulmonary epithelialcells induced by lipopolysaccharide （LPS）. Methods： Different concentrations of water extract and alcohol extract ofCitrus rnedica were applied to A549 cells. The effects of water extract and alcohol extract on the viability of A549 cellswere detected by CCK8 assay. The A549 cells were intervened with LPS（10 μg/mL） and the extracts. The contents ofIL - 8 and IL - 10 were detemfined in the supernatant by ELISA assay after 24 hours. Results ： There was no significantdifference in the effect of water extract and alcohol extract on the activity of A549 cells（P 〉 0.05 ）. After pulmonary epi-thelial cells were acted by lipopolysaccharide（LPS） , the release of IL -8 and IL -10 increased（P 〈0.01 ）. The waterextract and the alcohol extract could reduce the increase of IL- 8 release（P 〈 0.01 ）. The secretion of IL -10 was in-creased significantly by the water extract （P 〈 0.01 ） ; whereas it was inhibited by the alcohol extract （P 〈 0. 01 ）.Conclusion： The water extract and the alcohol extract of Citrus medica have no damage effect on pulmonary epithelialcells; they can inhibit the release of pro - inflammatory cytokines and increase the release of anti - inflammatory cyto-kines in acute lung injury model induced by lipopolysaccharide.
