摘 要:目的:探讨细胞间黏附分子-1(ICAM-1)在SD大鼠局灶性脑缺血后不同时相脑组织中的表达及八味沉香散对其影响。方法:雄性SD大鼠360只,随机分为假手术组、模型组、八味沉香散大剂量组(0. 8 g/kg)、八味沉香散中剂量组(0. 4 g/kg)和八味沉香散小剂量组(0. 2 g/kg)。每组随机分为1 h组、6 h组、12 h组、24 h组、72 h组、120 h组等6个时相组,每一时相12只大鼠。采用改良Zea Longa线栓法制备大脑中动脉(MCAO)栓塞模型,实时荧光定量(Real Time-PCR)法检测八味沉香散干预后对局灶性脑缺血大鼠不同时相脑组织ICAM-1表达的影响。结果:与假手术组比较,模型组神经功能评分提高,且模型组评分均> 2分,证明模型制作成功。ICAM-1在脑缺血损伤1 h后开始上升,于24 h达到高峰,72 h后逐渐下降(P <0. 05);八味沉香散各剂量组ICAM-1表达较模型组降低,但仍高于假手术组(P<0. 05)。结论:八味沉香散可能通过降低局灶性脑缺血大鼠脑组织中ICAM-1表达量来减轻神经功能损伤及炎症反应对脑组织的缺血性损伤,从而发挥对局灶性脑缺血损伤的治疗作用。Objective: To investigate the expression of intercellular adhesion molecule-1( ICAM-1) in different phases of brain after focal cerebral ischemia in SD rats and the effect of Bawei Chenxiang powder on it. Methods: 360 male SD rats were randomly divided into sham operation group,model group,high-dose group of Bawei Chenxiang powder( 0. 8 g/kg),middle-dose group of Bawei Chenxiang powder( 0. 4 g/kg) and Bawei incense scattered small dose group( 0. 2 g/kg).Each group was randomly divided into 1 h group,6 h group,12 h group,24 h group,72 h group,120 h group and 6 other phase groups. Each group had 12 rats. The modified middle cerebral artery( MCAO) embolization model was established by a modified Zea Longa method. Real-time fluorescence quantitative PCR was used to detect the expression of ICAM-1 at different phases in rats with focal cerebral ischemia after intervention of Bawei Chenxiang Powder. The effect of Bawei Chenxiang powder on the mRNA expression of ICAM-1 in brain tissue of rats with cerebral ischemia at different time points was analyzed. Results: Compared with the sham-operated group,the neurological function scores of the model group were significantly increased,and the model group scores were all more than 2 points,which proved the success of the model. ICAM-1 began to increase 1 hour after cerebral ischemic injury,reached a peak at 24 hours,and gradually decreased after 72 hours( P〈 0. 05); the expression of ICAM-1 in Bawei Chenxiang Powder was significantly lower in each dose group than in the model group.( P 〈 0. 05),but still higher than the sham group. Conclusion: Bawei Chenxiang Powder can reduce the ischemic injury of brain tissue by reducing the expression of vascular cell adhesion molecule-1( ICAM-1) in the brain tissue of rats with focal cerebral ischemia to play a role in the treatment of focal cerebral ischemic injury.
