摘 要:目的评价心搏骤停/心肺复苏大鼠脑损伤海马冷诱导RNA结合蛋白(CIRP)表达的变化。方法清洁级健康雄性sD大鼠72只,体重280—350g,8.10周龄,采用随机数字表法分为2组:假手术组(S组,n=18)和缺血再灌注组(I/R组,n=54)。S组只进行气管插管及动静脉穿刺;I/R组经食管电刺激诱发室颤建立心搏骤停模型。于复苏后12、24和48h时处死大鼠取海马组织。采用Westernblot法和qPCR法分别检测CIRP、TNF.仪和IL-1B及其mRNA的表达;光镜下观察海马病理学结果。结果与s组比较,I/R组复苏后24和48h时海马CIRPmRNA表达上调,复苏后12、24和48h时TNF-amRNA表达上调,复苏后12和24h时IL-1BmRNA表达上调,复苏后12、24和48h时CIRP、TNF-α和IL-1D表达均上调(P〈0.05)。I/R组海马CAl区出现病理学损伤。结论心搏骤停/心肺复苏大鼠脑损伤时海马CIRP表达上调,促进中枢炎症反应。[ Abstract] Objective To evaluate the changes in expression of coldinducible RNAbinding pro tein (CIRP) in hippocampus during brain injury in a rat model of cardiac arrestcardiopulmonary resuscita tion. Methods Seventytwo cleangrade healthy male SpragueDawley rats, weighing 280350 g, aged g10 weeks, were divided into 2 groups using a random number table: sham operation group (S group, n= 18) and ischemiareperfusion group (I/R group, n= 5,*). Tracheal intubation was performed and artery and veins were punctured in group S. Ventricular fibrillation was induced by transoesophageal cardiac pa cing to establish the model of cardiac arrest in group I/R. Rats were sacrificed at 12, 24 and 48 h after re suscitation and the hippocampus was harvested for determination of CIRP, tumor necrosis factoralpha (TNFa) and interleukinlbeta (ILI[3) protein and mRNA expression (by quantitative polymerase chain reaction or Western blot) and for determination of pathological changes of hippocampi (with a light micro scope). Results Compared with group S, the expression of CIRP mRNA in hippocampus was upregula ted at 24 and 48 h after resuscitation, the expression of TNFct mRNA was upregulated at 12, 24 and 48 h after resuscitation, the expression of ILI[3 mRNA was upregulated at 12 and 24 h after resuscitation, andthe expression of CIRP, TNFct and IL113 was upregulated at 12, 24 and 48 h after resuscitation in group I/R (P〈0. 05). Pathological changes in hippocampal CAI region were found in group I/R. Conclusion The expression of CIRP in hippocampus is upregulated, which promotes central inflammatory responses during brain injury in a rat model of cardiac arrestcardiopulmonary resuscitation.
