摘 要:目的探讨芦丁对肾间质纤维化影响及作用机制。方法采用单侧输尿管梗阻(unilateral uretaral obstruction,UUO)建立肾间质纤维化模型,将雄性Wistar大鼠随机分为4组,每组6只:假手术组、假手术组+芦丁治疗组、模型组、模型组+芦丁治疗组。术后24 h开始,芦丁治疗组大鼠予芦丁[100mg/(kg·d)溶于1%羧甲基纤维素钠(CMC-Na)溶液]灌胃,其余两组大鼠予等体积1%CMC-Na溶液灌胃。术后2周处死各组大鼠,取梗阻侧肾组织分别用HE、天狼星红(Sirius red)染色评估各组大鼠肾间质损伤指数和胶原沉积,免疫组化法检测肾组织内胶原蛋白Ⅰ(collagenⅠ,Col-Ⅰ)和纤维连接蛋白(fibronectin,FN)的表达水平。Western Blot方法检测转化生长因子β1(TGF-β1)、磷酸化Smad3(p-Smad3)的蛋白表达水平。结果模型组大鼠肾间质损伤及纤维化程度均高于假手术组,肾组织内Col-Ⅰ、FN、TGF-β1、pSmad3蛋白表达水平均明显增高。与模型组比较,芦丁治疗组肾间质损伤及纤维化程度明显减轻,肾组织内Col-Ⅰ、FN、TGF-β1、p-Smad3蛋白表达水平均明显减少。结论芦丁具有改善UUO所致大鼠肾间质纤维化的作用,其作用可能与抑制TGF-β1/Smad3信号通路有关。Objective To investigate the effect and involved mechanism of rutin on tubulointerstitial fibrosis in unilateral ureteral obstruction (UUO) rat models. Methods UUO rat models were used. Male Wistar rats were randomly divided into four groups with 6 rats each :including vehicle - or rutin - treated sham operated groups, and vehicle - or rutin - treated UUO groups. From the first day after initial UUO, Rats of rutin - treated groups were received daily oral gavage of rutin( 100 mg/kg) dissolved in 1% carboxymethylcellulose - Na ( CMC - Na) buffer for 2 weeks. Equal volume of CMC - Na buffer was used as a vehicle. All rats were sacrificed 2 weeks after surgery. Tubulointerstitial injury index, interstitial collagen deposition were evaluated by HE and Sirius red staining. Expression of collagenⅠ ( Col -Ⅰ ) and fibronectin (FN) in renal tissues were analyzed through immunohistochemistry. Western blot was used to detect the protein expression level of TGF - β1 and p - Smad3. Results Compared with sham operation groups, the tubulointerstitial injury index and interstitial collagen deposition in UUO groups were significantly increased. The expression of Col- Ⅰ , FN,TGF- β1 and p- Smad3 were also increased. However, these morphological changes and excessive collagen deposition were notably reduced after treatment with rutin. Meanwhile, the expression of Col - Ⅰ ,FN ,TGF - β1 and p - Smad3 were also decreased. Conclusion The effect of rutin on ameliorating tubulointerstitial fibrosis in UUO rats may be correlated with its inhibition on TGF - β1/Smad3 signal pathway.
