日本龟蜡蚧雌成虫越冬前后4种代谢酶和3种保护酶活力变化研究

摘  要:采用分光光度计法测定和比较了越冬前(2015年10月)、越冬期(2016年1月)和越冬后(2016年4月)日本龟蜡蚧雌成虫体内4种代谢酶(海藻糖酶、山梨醇脱氢酶、淀粉酶、蔗糖酶)以及3种保护酶(超氧化物歧化酶、过氧化氢酶、过氧化物酶)的活力。结果表明,在越冬前后期间,4种代谢酶和3种保护酶的酶活均有显著差异(P<0.05)。其中,海藻糖酶和山梨醇脱氢酶比活力在越冬期最高,分别为22.707 8×10^(-9)mol/s和24.816 6×10^(-9)mol/s,随着越冬时间的延长呈下降趋势;淀粉酶在越冬期和越冬后无显著差异性,而均与越冬前差异显著(P<0.05),淀粉酶在越冬前期比活力活力最高,为4.354 2×10^(-9) mol/(g·s),越冬期最低,在越冬结束后,淀粉酶的比活力开始缓慢上升;蔗糖酶在越冬期的比活力最高,为2.753 8×10^(-9))mol/(g·s),而在翌年4月份越冬结束后开始取食和活动,比活力明显比10月份要高。超氧化物歧化酶在整个越冬期间均呈现差异显著性,在越冬后酶活力最低(781×10^(-9)mol/s);过氧化氢酶活力在越冬后期,随时间的变化而急剧下降,越冬前最高(794×10^(-9)mol/s),越冬后最低(227×10^(-9)mol/s);过氧化物酶在越冬前与越冬期无显著差异,但均与越冬后差异显著(P<0.05),在越冬前酶活力最高(402×10^(-9)mol/s),在越冬后最低(150×10^(-9)mol/s)。Activities of four kinds of me tabolic enzym es(trehalase, sorbitol dehydrogenase, amylase and invertase) and three kinds of protective enzymes(SOD, POD and CAT) in female adults of Ceroplastes japonicus before overwintering(October, 2015), during overwintering(January, 2016) and after overwintering(April, 2016) were measured by spectrophotometer method. The results showed that activities of metabolic and protective enzymes had significantly differences in different overwintering stages(P﹤0.05). Among them, activity of trehalase [22.7078×10^-9 mol/(g·s)] and SDH [24.8166 ×10^-9 mol/(g·s)] were highest during overwintering period, then gradually decreased with prolongation of overwintering time; amylase activity had no significant difference between overwintering period and after-overwintering period, but had significantly difference from that of pre-overwintering period. Specific activity of amylase was highest[4.354 2 ×10^-9 mol/(g·s)] in pre-overwintering period, lowest in overwintering period, and began to rise slowly after overwintering. Specific activity of invertase was the highest [2.753 8 ×10^-9 mol/(g·s)] during overwintering. The insect began to feed and act after overwintering in April of the following year, and specific activity of invertase was significantly higher than that in last October. Specific activity of SOD showed significant difference among overwintering, pre-overwintering and after-overwintering periods, and it was lowest after overwintering(781 ×10^-9 mol/s). Activity of CAT decreased significantly with time going, and it was highest before overwintering(794 ×10^-9 mol/s) and lowest after overwintering(227×10^-9 mol/s). Activity of POD before and during the overwintering period was not significantly different,while both of them had significant differences with that of after-overwintering period, as it was highest before overwintering(402×10^-9 mol/s) and lowest after overwintering(150×10^-9 mol/s).
