摘 要:目的:通过观察香砂六君丸对脾虚痰浊大鼠小肠黏膜细胞JAK2/STAT3通路的影响,探讨健脾化痰法的生物学机制。方法:40只雄性SD大鼠,随机分为对照组、脾虚痰浊组(模型组)、香砂六君丸低剂量治疗组(香低组)、香砂六君丸中剂量治疗组(香中组)和香砂六君丸高剂量治疗组(香高组),每组8只;全自动生化仪检测血脂;ELISA法检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)和脂多糖(LPS)水平及谷胱甘肽过氧化物酶(GSH-Px)活性;免疫组化法检测小肠黏膜磷酸化(p)-JAK2/STAT3和Bcl-2表达。结果:与对照组比较,模型组甘油三酯(TG)、低密度脂蛋白(LDL)、MDA和LPS水平及p-JAK2/STAT3表达上升,高密度脂蛋白(HDL)和SOD水平、GSH-Px活性以及Bcl-2表达下降,均有统计学差异。与模型组比较,后3组的TG、LDL和LPS水平及p-JAK2表达下降,且SOD水平和GSH-Px活性上升;后两组的MDA水平减少;香高组HDL水平和Bcl-2表达增加,p-STAT3表达下降,均有统计学意义。与香低组比较,后两组MDA浓度下降,均有统计学差异。结论:健脾化痰法的生物学机制可能涉及调控血脂、抗氧化、抑制内毒素过度释放和增强小肠黏膜细胞抗凋亡能力等。Objective: To study the effect of Xiangsha Liujun Pill(XLP) on small intestinal(SI) mucosal JAK2/STAT3 pathway in spleen deficiency and phlegm turbid rats,attempting to explore the biological basis of the principle of invigorating spleen and resolving phlegm. Methods: Forty male SD rats were randomly divided into the control group,spleen deficiency and phlegm turbid group(model group),XLP low-dose treating group(XLG),XLP middle-dose treating group(XMG) and XLP high-dose treating group(XHG),8 rats in each group. Automatic chemistry analyzer was used to measure serum lipid levels. ELISA was employed to detect serum levels of superoxide dismutase(SOD),malondialdehyde(MDA) and lipopolysaccharide(LPS) and glutathion peroxidase(GSH-Px) activity. The immunohistochemistry was used to detect expressions of phosphorylated(p)-JAK2,p-STAT3 and Bcl-2 in SI muscosa. Results: Compared with those in the control,both total glyceride(TG),low density lipoprotein(LDL),MDA and LPS levels and p-JAK2/STAT3 expressions were increased significantly,and high density lipoprotein(LDL) and SOD level,GSH-Px activitability and Bcl-2 expression were decreased markedly in the model groups. Compared with those in the model groups,but TG,LDL and LPS levels and p-JAK2 expression were lowered,and SOD level,GHS-Px activitability and Bcl-2 expression were increased,significantly,in later 3 groups. MDA levels in later 2 groups were significantly reduced. HDL level and Bcl-2 expression were increased,and p-STAT3 expression was reduced in XHG obviously. Compared with that in XLG,MDA levels in later 2 groups were decreased significantly. Conclusion: The biological base of the principle of invigorating spleen and resolving phlegm might relate to control serum lipid,opposite oxidative stress,inhibit endotoxin overrelease and increase SI mucosal anti-apoptotic ability.
