山核桃叶总黄酮干预去势SD大鼠脂肪组织异位沉积作用研究  

Investigation of the Effect of Total Flavonoids in Carya Cathayensis Leaf on Adipose Tissue Ectopic Deposition in Deovary SD Rats

摘  要:[目的]研究山核桃叶总黄酮对去势SD大鼠脂肪组织异位沉积的干预作用。[方法] 60只SD大鼠分为模型组、假手术组、雌二醇组、山核桃叶总黄酮低、中、高剂量组,每组10只。除假手术组外,其余各组均摘除卵巢建立去势模型,模型组和假手术组予0.9%氯化钠注射液灌胃,雌二醇组给予雌二醇1mg/kg灌胃,山核桃叶总黄酮低、中、高剂量组分别予山核桃叶总黄酮50mg/kg、100mg/kg、200mg/kg灌胃。各组大鼠灌胃1次/d,容积0.3~0.4mL/只,共2个月。给药结束后处死大鼠,计算腹部脂肪系数、主动脉弓脂肪系数;制作肝脏组织、主动脉弓血管、腹部脂肪组织切片,HE染色后镜下观察;酶标比色法检测血清总胆固醇(total cholestero,TC)、甘油三酯(triglyceride,TG)、肝脏TG、肝脏游离脂肪酸(free fatty acid,FFA)的水平;Q-PCR法检测过氧化物酶体增殖体激活受体γ(peroxisome proliferator activated receptorγ, PPARγ)m RNA表达量,Western blot检测其蛋白表达量。[结果]与模型组比较,山核桃叶总黄酮高剂量组能显著降低去势大鼠血清TG水平(P<0.01),下调肝脏TG水平(P<0.05),但肝脏FFA水平无明显变化(P>0.05);与模型组比较,雌二醇组和山核桃叶总黄酮各剂量组肝脏细胞内脂滴数量减少,主动脉弓处、腹部脂肪细胞直径减小;与模型组比较,山核桃叶总黄酮中、高剂量组PPARγ蛋白表达下调(P<0.01),中剂量组PPARγmRNA表达下调(P<0.05)。[结论]山核桃叶总黄酮可改善去势SD大鼠脂肪在肝脏、主动脉弓血管周围组织的异位沉积,其作用机制可能与调节PPARγ表达,减少脂肪组织分化,减少FFA释放,降低血脂水平相关。[Objective]To study the effect of total flavonoids in Carya cathayensis leaf on the ectopic deposition of adipose tissue in deovary SD rats. [Methods] Sixty SD rats were divided into model group, sham operation group, estradiol group, low dose, medium dose, and high dose group of total flavonoids in Carya cathayensis leaf, with 10 rats in each group. In addition to sham operation group, all other groups were ovariectomized to establish castration model, model group and sham operation group were administrated with 0.9% sodium chloride injection, estradiol group was given with estradiol 1mg/kg, low dose, medium dose and high dose group was given total flavonoids in Carya cathayertsis leaf 50mg/kg, 100mg/kg, and 200mg/kg, respectively. The volume of garage was controlled at 0.3 to 0.4mL/dose, once a day for 2 months. After the administration, the tats were executed, and the abdominal fat coefficient and the aortic arch fat coefficient were calculated. To make liver tissue, aortic arch vessels, abdominal adipose tissue HE section observation; detection of serum total cholesterol(TC), serum triglyceride TG(TG) and liver free fatty acids(FFA) by enzyme eolorimetric eolorimetric; the expression of peroxisome proliferator activated receptor γ (PPARγ) mRNA was detected by Q-PCR and Western blot was used to detect the expression of PPARγ mRNA and protein, [Results] Compared with model group, high dose group of total flavonoids in Carya cathayerysis leaf significantly decreased the serum TG levels in ovariectomized rats(P〈0.01) and down-regulated the liver TG levels(P〈0.05), but there was no significant change in liver FFA(P〉0.05). Compared with model group, the number of lipid droplets in liver cells in estradiol group and groups of total flavonoids in Carya cathayertsis leaf decreased, and the diameter of adipocytes at the aortic arch and abdomen decreased. Compared with model group, the expression of PPARγ, protein in medium and high dose groups was down-regulated(P〈0.01), and
