摘 要:目的通过分析癌毒清颗粒(AG)干预二乙基亚硝胺(DEN)诱导SD大鼠肝癌(HCC)模型在不同时间肝脏病理,肝组织细胞凋亡相关基因(Bcl-2、Bax、Fas/Fasl、Casepase-3、Survivin)的表达,揭示AG防治原发性肝癌的机制。方法 40只雄性SD大鼠,随机均分为预防组和对照组,两组均饲以64 ppm浓度的DEN水溶液16周诱癌,然后饲以纯净水。预防组予AG 0.2 g/100 g灌胃,用药6 d,停药1 d,称重调整用药剂量,干预18周。两组在第4、8、12、16、18周分别随机处死4只大鼠,剖腹取出肝脏,选取阳性部位,HE染色,用于病理研究。PCR检测Bcl-2、Bax、Fas/Fasl、Casepase-3、Survivin基因表达。结果预防组各时间点均高表达Caspase-3 mRNA,低表达Bcl-2 mRNA;Fas/Fasl mRNA在第4周显著表达;Bax mRNA第12周显著表达;Survivin mRNA第8周表达下调。结论 AG干预模型大鼠是通过促进Bax mRNA、Caspase-3 mRNA、Fas mRNA表达来实现促进受损细胞、HCC细胞凋亡。另外AG干预后细胞凋亡相关基因表达在各时间点存在不一致性,提示AG防治HCC是通过多种细胞凋亡途径来实现的。Objective To explore the mechanism of aiduqing granules in the prevention and treatment of primary liver cancer through analyzing the live pathology at different time points and the expression of the apoptosis-related genes(Bcl-2,Bax,Fas/Fasl,Casepase-3 and Survivin) in SD rat models of diethylnitrosamine(DEN)-induced liver cancer.Methods A total of 40 male rats were randomized into a prevention group and a control group. The cancer models were induced with DEN solution,at the concentration of 64 ppm for 16 weeks and fed with purified water. In the prevention group,the lavage with aiduqing granules,0. 2 g/100 g was given for 6 days. After the discontinuity for 1 d,the dose was adjusted on base of weight,totally for 18 weeks. In the week 4,8,12,16 and 18,4 rats were sacrificed randomly. The positive tissue of the liver was collected for pathological study with HE staining. PCR was adopted to determine the expressions of Bcl-2,Bax,Fas/Fasl,Casepase-3 and Survivin. Results In the prevention group,Caspase-3 mRNA was highly expressed and Bcl-2 mRNA less expressed at each time point; Fas/Fasl mRNA was significantly expressed in the week 4,Bax mRNA was significantly expressed in the week 12 and Survivin mRNA expression was down-regulated in the week 8.Conclusion The intervention with aiduqing granules contributes to the apoptosis of the damaged cells and HCC cell through promoting the expressions of Bax mRNA,Caspase-3 and Fas mRNA. After the intervention,the apoptosis-induced genes were expressed differently at each time point. It is suggested the prevention and treatment of liver cancer with aiduqing granules is achieved through the apoptosis of different cells.
