摘  要:2010年以来,猪流行性腹泻给我国养猪业造成了严重的经济损失。为分析PEDV在我国最新的遗传变异情况,本研究对2014-2016年江苏地区20株PEDV野毒株的S基因进行克隆和测序,并对其遗传进化、序列变化、抗原表位等情况进行分析。遗传进化树显示,所测的PEDV毒株可分为G1、G2两大分支,12株流行野毒株位于G2b分支上,8株属于G1群S-Indel型,而与疫苗毒株亲缘关系较远。序列对比发现,氨基酸变化主要集中于S1区N端,所测毒株JSCZ1601出现了特异性的缺失。20株毒株的核苷酸(氨基酸)同源性为95. 1%~99. 8%(94. 6%~99. 9%),与我国流行毒株AH2012、疫苗毒株Attenuated DR13、欧美毒株CV777的同源性为95. 5%~99. 3%(94. 6%~99. 0%)、93. 5%~96. 0%(92. 5%~96. 3%)、93. 6%~95. 8%(93. 0%~96. 5%)。表位分析表明,主要变异位于COE、SS2区域,两个线性表位(SS6和against2C10)相对保守;跨膜螺旋区域的预测得知毒株之间存在部分差异。这些结果表明,江苏省流行的PEDV毒株发生较明显的变异,需研发新的疫苗来控制PEDV的暴发。Since 2010, swine epidemic diarrhea has caused serious economic losses to pig industry in our country. In order to analyze the latest genetic variation of PEDV in China,we cloned and sequenced the S gene of 20 PEDV wild strains in Jiangsu prov ince from 2014 to 2016, and analyzed its genetic evolution, sequence changes and epitopes. The phylogenetic tree showed that the phylogenetic tree could be divided into two major branches, G1 and G2. Twelve endemic strains were located on the G2b branch. All the eight strains of G1 belonged to S-Indel type, but had a distant relationship with the vaccine strain. Sequence comparison found that amino acid changes were mainly concentrated in the amino terminal S1 region, and the measured strain ]SCZ1601 showed a spe- cific deletion. The nucleotide (amino acid) homology of the 20 strains was 95.1% -99.8% (94.6% -99.9% ). The homology with the strain AH2012, Attenuated DR13 and CV777. 99.3% (94.6% - 99.0% ), 93.5% - 96.0% (92.5% - 96.3% ), and 93.6% -95.8% (93.0% -96.5% ),respectirely. Analysis of epitopes showed that the major variations were located in the COE and SS2 regions, and the two linear epitopes ( SS6 and against 2C10) were relatively conserved. The prediction of the transmembrane helix region revealed partial differences between the strains. The prevalence of PEDV strains in Jiangsu province has been changed significantly and new vaccines should be developed to control the outbreak of PEDV.
