摘 要:目的:探讨地佐辛对大鼠皮层星形胶质细胞的影响。方法:选取SD大鼠的乳鼠10只,提取大鼠皮层星形胶质细胞进行原代培养,按照给予大鼠地佐辛剂量的不同,将其分为对照组、低剂量组、中剂量组和高剂量组,其中低剂量组、中剂量组和高剂量组分别给予地佐辛3 g、6 g和12 g,对照组给予等量生理盐水。对4组大鼠采用四甲基偶氮唑(MTT)比色法检测细胞活性;免疫荧光法检测胶质纤维酸性蛋白(GFAP)表达;细胞划痕实验检测细胞迁移情况。比较4组大鼠细胞活性光密度(OD)值、GFAP表达和细胞迁移情况。结果:对照组细胞活性OD值、GFAP荧光强度和迁移细胞数分别为(0.654±0.110)、(18.122±1.241)和(30.24±3.55)个,明显高于低剂量组、中剂量组和高剂量组;高剂量组细胞活性OD值、GFAP荧光强度和迁移细胞数分别为(0.357±0.108)、(5.249±0.977)和(3.20±1.03)个,明显低于低剂量组和中剂量组;中剂量组细胞活性OD值、GFAP荧光强度和迁移细胞数分别为(0.654±0.110)、(10.528±1.311)和(13.41±4.10)个,明显低于低剂量组;对照组细胞活性OD值、GFAP荧光强度和迁移细胞数与其他3组比较,其差异有统计学意义(F=12.144,F=84.645,F=110.467;P<0.05)。结论:地佐辛可抑制大鼠皮层星形胶质细胞活性及迁移,下调GFAP的表达,值得进一步研究。Objective To investigate the effect of Dezocine on cortical astrocytes of rat. Methods: 10 SD rats were selected to extract their cortical astrocytes for primary culture. And they were randomly divided into different groups with various dose of dezocine (the control group, the low dose group, the middle dose group and the high dose group). And the low dose group, middle dose group and high dose group were given 3g, 6g and 12 g dezocine, respectively, and the control group was given the same volume of normal saline. The cell activities of SD rats of 4 groups were detected by MTT colorimetric method, and the expression of glial fibrillary acidic protein (GFAP) was detected by immunofluorescence method, and the cell migration was detected by the cell wound scratch assay. The absorbance of cell activity, GFAP expression and cell migration were compared among the 4 groups, respectively. Results: The absorbance value of cells activation, the fluorescence intensity of GFAP and the number of migrated cells of control group were (0.654±0.110), (18.122±1.241) and (30.24±3.55), respectively, and they were significantly higher than that of low dose group, middle dose group and high dose group. The absorbance value of cells activation, the fluorescence intensity of GFAP and the number of migrated cells of high dose group were (0.357±0.108), (5.249±0.977) and (3.20±1.03), respectively, and they were significantly lower than that of low dose group and middle dose group. The absorbance value of cells activation, the fluorescence intensity of GFAP and the number of migrated cells of middle dose group were (0.654±0.110), (10.528±1.311) and (13.41±4.10), respectively, and they were significantly lower than that of low dose group. The differences of absorbance value, fluorescence intensity of GFAP and the number of migrated cells between the control group and the other three groups were significant (F=12.144, F=84.645, F= 110.467, P〈0.05).
