摘  要:目的采用TK基因突变试验研究卷烟烟气的遗传毒性。方法以L5178Y tk+/-3. 7. 2C小鼠淋巴瘤细胞为研究对象,3R4F参比卷烟的烟气为受试物,卷烟烟气经DMSO萃取后,设置试验剂量组为20、40、80、100、150μg/m L,另设二甲基亚砜溶剂对照组和环磷酰胺阳性对照组,对L5178Y tk+/-3. 7. 2C小鼠淋巴瘤细胞染毒处理及表达。结果在加S9的情况下,80μg/m L的突变频率是溶剂对照组3倍以上,实验结果呈阳性。结论在此次试验条件下,卷烟烟气具有TK基因致突变作用。Objective To investigate the potential genotoxicity of cigarette smoke using the TK gene mutation test. Methods The cigarette smoke condensate ( CSC) of the reference cigarette 3R4F was extracted by DMSO. The L5178Y tk^ + / -3, 7, 2C mousely mphoma cells were treated with CSC at different concentrations, 20、40、80、100 and 150 μg / mL, for 3 h. Dimethyl sulfoxide was the solvent control group, and cyclophosphamide was positive control group. Results With metabolic activation, the TK gene mutation frequency of 80 μ g / mL group was more than 3 times higher than that of the solvent control group. Conclusion The results indicatethat cigarette smoke containing toxicant inducing TK gene mutation.
