摘 要:本文探讨利用超声波辅助乙醇提取法提取,AB-8大孔吸附树脂纯化的东风菜总黄酮对四氧嘧啶糖尿病小鼠降血糖、降血脂及抗氧化作用。将小鼠随机分为正常组、模型组、二甲双胍组及东风菜总黄酮50 mg/kg、100 mg/kg、200 mg/kg剂量组,连续4周灌胃给药,记录其进水量、进食量及体重的变化,末次给药后尾静脉取血测定血糖、糖耐量、血脂指标;分离肝组织测定抗氧化指标。试验结果表明:东风菜总黄酮能降低小鼠空腹血糖值(Glu),改善糖耐量,并有效缓解其"三多一少"症状,其中200 mg/kg剂量组降糖效果与阳性组作用效果相当;能有效调节其血清中TC、TG、LDL-C、HDL-C浓度,其中最佳试验组较模型组的HDL-C值提高了44.55%,TG降低了18.71%,TC值降低了27.38%,LDL-C值降低了24.81%,表现出良好的降血脂作用;能降低其肝组织中MDA的含量,提高GSH-Px、CAT、SOD的含量,增强其抗氧化能力,其中最佳试验组较模型组的MDA值降低18.79%,GSH-Px值提高了41.02%,SOD值提高了15.40%,CAT值提高了47.84%,具有良好的抗氧化作用。作用机制可能与其提高小鼠抗氧化能力、改善血脂代谢紊乱等有关。The effects of Doellingeria scaber total flavonoids on the hypoglycemia, blood lipid and oxidation in diabetic mice were investigated. Alloxan-iduced diabetic mice were fed with Doellingeria scaber total flavonoids, flavonoids and polysaccharide at the dose of 50 mg/kg, 100 mg/kg, 200 mg/kg, respectively, for 4 weeks. The amount of water, food intake and body weight changes were recorded. The results showed that the total flavonoids can reduce the fasting blood glucose(Glu), improve the glucose tolerance, and effectively relieve the "a little" sanduo symptoms. The total flavonoids can also effectively regulate the serum TC, TG, LDL-C, HDL-C concentrations. As compared with model group, the HDL-C value of the experimental group increased by 44.6%, while the TG, TC and LDL-C values were reduced by 18.7%, 27.4% and 24.8%, respectively. Furthermore, the total flavonoids can reduce the content of MDA in liver tissue, increase the content of gsh-px and CAT, SOD, enhance its antioxidant capacity. These effects may be associated with improved antioxidant capacity and improved lipid metabolism in diabetic mice.
