摘 要:目的:探讨电针刺激对慢性不可预见性应激(CUS)大鼠抑郁行为的改善作用及内源性大麻素受体1(CB1)、二酰基甘油脂肪酶(DAGLα)、脂肪酰胺水解酶(FAAH)和单酰基甘油脂肪酶(MAGL)基因表达的影响。方法:24只SD大鼠随机分为对照组(Sham),模型组(CUS),模型+电针治疗组(CUS+EA),每组8只。CUS组以及EA+CUS组大鼠接受慢性不可预见性刺激处理(CUS)造模; EA+CUS组在造模第14 d开始给予电针干预,每天给予百会穴电针刺激30 min,电流1 m A,频率2/15 Hz疎密波,连续7 d; Sham组和CUS组给予假刺激;静养两周后,通过旷场,强迫游泳以及糖水偏好实验检测各组大鼠的抑郁样行为。行为学检测结束后处死大鼠,通过real time RT-PCR检测海马CB1受体、DAGLα、FAAH及MAGL的mRNA表达情况。结果:(1) CUS处理可以导致大鼠明显的抑郁样行为,包括进入旷场中心区次数和时间减少(P <0. 05),强迫游泳不动时间增加(P <0. 01),糖水摄取量下降(P <0. 05)。CUS组大鼠海马的CB1、DAGLα的mRNA表达下调(P <0. 05),而FAAH和MAGL表达上调(P <0. 05)。(2)电针刺激可以缓解CUS大鼠的抑郁样行为,CUS组与EA+CUS组之间存在显著性差异(P <0. 05)。(3)电针刺激可以恢复CUS大鼠海马的内源性大麻素相关基因表达水平。结论:电针刺激可以缓解CUS模型大鼠的抑郁样行为,调节抑郁模型大鼠海马的CB1受体和内源性大麻素代谢酶基因表达。Objective: To investigate the effect of electroacupuncture( EA) stimulation on the improvement of depression and the expression of endocannabinoid system-related genes,including cannabinoid receptor1( CB1),diacylglycerol( DAGLα),fatty acid amide hydrolase( FAAH) and monoacylglycerol( MAGL) in hippocampus in CUS rats.Methods: Twenty four SD rats were randomly assigned to the following 3 groups( n = 8 for each group) : sham,CUS,EA + CUS.Rats in the CUS and EA + CUS groups were subjected to CUS.Then,the EA + CUS group received EA stimulation at"Baihuixue"at frequency of 2/15 Hz for 30 min daily for seven days post-CUS.Sham and CUS rats received sham stimulation.After 14 days of EA treatment,rats experienced open field test,forced swim test and sucrose preference test,and then sacrificed and the mRNA level of CB1,DAGLα,MAGL and FAAH in the hippocampus were determined by real-time PCR.Results:( 1) CUS induced depression-like behavior.Rats in CUS group showedsignificant less time in the central field( P〈0.05),decreased sucrose preference and increased immobility time in forced swim test( P〈0.05),simultaneously mRNA level of CB1 and DAGLα in hippocampus were decreased( P〈0.05),while MAGL and FAAH were increased( P〈0.05);( 2) Electroacupuncture stimulation reversed depressivelike behaviors in rats exposed to CUS paradigm as compared between CUS and EA + CUS groups;( 3) Electroacupuncture stimulation restored the mRNA level of endocannabinoid system-related genes in the hippocampus of CUS rats.Conclusion: Electroacupuncture stimulation can ameliorate depressive-like behaviors in rats exposed to CUS and restore the the mRNA level of endocannabinoid system-related genes in the hippocampus.
