摘 要:血脑屏障通透性增加与血管性认知功能障碍及血管性痴呆的发病和进展关系密切。紧密连接是位于脑微血管内皮细胞顶端的蛋白分子复合体,是调节血脑屏障通透性的重要结构和功能基础。各种血管性致病因素作用于紧密连接蛋白,通过影响后者的表达,结构和功能造成血脑屏障的破坏,这是血管性认知障碍的重要发病机制之一。本文就血脑屏障紧密连接复合体的分子构成、功能及其在血管性认知障碍中的表达变化、信号通路机制的研究进展做一综述,为血管性认知障碍的临床防治提供一个全新的思路。The increased permeability of blood - brain barrier is closely related to the pathogenesis and progression of vascular cognitive impairment and vascular dementia. Tight junction is the most apical protein molecular complex within the intercellular cleft, which is an important structure and functional basis for regulating the permeability of blood - brain barrier. Various vascular pathogenic factors disrupt blood - brain - barrier by influencing the expression, structure and function of tight junctions, which is one of the important pathogenesis of vascular cognitive impairment. This article reviews the molecular structure and function of the blood - brain barrier tight junction complex, its molecular structure, expression, functional role and signaling transduction mechanism in vascular cognitive impairment, providing a new perspective for the clinical prevention and treatment of vascular cognitive impairment.
