摘 要:猪精子对低温尤为敏感,极易遭受低温损伤。本文旨在探讨冷冻前后猪精子超微结构变化,为改善猪精子冷冻保存方法奠定基础。通过手握法采集姜曲海公猪精液,使用0.25 m L细管对精液进行超低温冷冻保存,用扫描电镜和透射电镜对冷冻前后精子超微结构进行观察。结果显示,冻后姜曲海猪精子活力、质膜完整率、线粒体活性和顶体完整率(分别为38.6%、40.5%、42.7%和43.6%)均显著(P〈0.05)低于新鲜精子(分别为81.9%、85.7%、89.5%和90.2%)。姜曲海猪精子全长约54.4 m,由头、颈、体和尾4部分组成;冻后正常的精子,表现为质膜、顶体和线粒体结构完整,顶体光滑,线粒体排列整齐,核染色质均匀;冻后异常精子主要表现为颈部断裂或膨胀,顶体表面粗糙或缺失,质膜膨胀或缺失,或线粒体移位。上述研究结果表明,超低温保存对姜曲海猪精子超微结构造成的损伤主要集中于质膜、顶体和线粒体。Because of the more sensitive to low temperature,boar sperm is highlysusceptible to the damage of low temperature.For improving the freezing efficiency,this study explored the ultrastructural changes of boar sperm before and after freezing.Jiangquhai boar semen was collected with hand holding method and cryopreserved with 0.25 m L straw.The ultrastructure of fresh or freezed-thawed sperms were observed with scanning electron microscope(SEM) and transmission electron microscope(TEM),respectively.In result,motility,membrane integrity rate,mitochondrial activity and acrosome integrity rate of the freezing group(38.6%,40.5%,42.7% and 43.6%,respectively) were significantly lower than those of the fresh group(81.9 %,85.7 %,89.5 % and 90.2 %,respectively).Jiangquhai boar sperm was about 54.4 μm in length,and was composed of head,neck,body and tail.Normal sperm after freezing was with continuous membrane,smooth acrosome,intact and aligned mitochondria and homogeneous chromatin. Abnormal sperm after freezing often showed different degrees of damage,such as ruptured or separated necks,rough or lost acrosome,expanded or lost membrane,or dislocated mitochondria.In conclusion,the cryodamage of Jiangquhai boar sperms after ultrastructural observation mainly appeared in the membrane,acrosome or mitochondria.
