摘  要:目的测定秦皮枝皮和干皮中秦皮甲素与秦皮乙素的含量,为秦皮规格等级的制定提供依据。方法采用Hypersil GOLD C_(18)色谱柱,乙腈-0.1%磷酸溶液(8∶92)为流动相,等度洗脱,检测波长为334 nm,流速为1.0 m L·min^(-1)。结果秦皮甲素、秦皮乙素在35 min内得到良好的分离,线性范围分别为28~560μg(r=0.9992)、1.98~39.6μg(r=0.9995)。样品测量结果显示,枝皮中秦皮甲素和秦皮乙素的总含量较干皮高。结论本文建立的方法适宜于秦皮中秦皮甲素与秦皮乙素的含量测定,结果能够为秦皮药材的合理应用、质量控制及商品规格新标准的制定奠定基础。Objective To determine the content of aesculin and esculetin in the branch skin and dry skin of Fraxini Cortex, and provide reference for the different commercial grades of Fraxini Cortex. Methods HypersilGOLD Ct8 was used, methanol-0.1% phosphoric acid as the mobile phase with gradient elution. The detection wavelength was 334 nm; the flow rate was 1.0 mL · min - 1. Results Aesculin and esculetin were separated at 35 min and showed good linearity at 28 -- 560μg (r = 0.9992) and 1.98 -- 39.6 μg (r = 0.9995), respectively. The total contents of aesculin and esculetin in the branch skin were higher than those in the dry bark. Conclusion HPLC can simultaneously determine the contents of aesculin and esculetin, which can be used for basis of rational application, quality control and standards of Fraxini Cortex.
