摘  要:目的:探讨化瘀消癥杀胚中药复方对输卵管妊娠滋养细胞凋亡及凋亡通路的影响。方法:(1)Hoechst染色法观察中药复方对输卵管妊娠滋养细胞形态学的影响;(2)透射电镜观察中药复方对输卵管妊娠滋养细胞超微结构的影响;(3)流式细胞术检测药物干预后输卵管妊娠滋养细胞的凋亡率;(4)采用Wester blot法及QT-PCR法检测Bax、Fasl、Caspase-3、Caspase-8、Caspase-9凋亡相关蛋白及基因的表达。结果:与阴性对照组比较,中药中、高剂量组凋亡率明显增加(P<0.01);中药高剂量组和中药+甲氨蝶呤组Bax、Fasl、Caspase-3、Caspase-9蛋白表达增加(P<0.01,P<0.05),中药中剂量组FasL、Caspase-3蛋白表达增加(P<0.05),中药低剂量组Bax、Caspase-3、Caspase-9蛋白表达增加(P<0.01,P<0.05),且中药高剂量组、中药+甲氨蝶呤组还可增加FasL、Caspase-3、Caspase-8基因表达(P<0.01,P<0.05)。结论:化瘀消癥杀胚中药复方可能通过诱导Caspase介导的死亡受体及线粒体凋亡通路而促进滋养细胞凋亡。Objective: To explore the effects of Chinese medicine compound of resolving stasis and abdominal mass embryo-killing on the apoptosis of tubal pregnancy nourishes and apoptotic pathway. Methods: ①The morphology of tubal pregnancy nourishes cells were observed by Chinese medicine compound through Hoechst staining. ②he effects of Chinese medicine compound on the ultrastructure of tubal pregnancy nourishes cells were observed by transmission electron microscope. @Flow cytometry was used to detect the apoptosis rate of tubal pregnancy nourishes cells. @The expression of apoptosis proteins and genes of Bax, Fasl, Caspase-3, Caspase-8 and Caspase-9 apoptosis were detected by Western blot and QT-PCR method. Results: The apoptotic rate of Chinese medicine in medium and high dose groups increased significantly (P〈0.01). The protein expressions of Bax, FasL, Caspase-3, Caspase-9 in high-dose of traditional Chinese medicine (TCM) group and TCM +methotrexate group were increased (P〈0.01, 1~〈0.05). The protein expressions of Fasl, Caspase-3 in middle-dose of TCM group were increased (P〈0.05). The protein expressions of Bax, Caspase-3, Caspase-9 in low-close of TCM group were increased (P〈0.01, P〈0.05). The gene expressions of FasL, Caspase-3, Caspase-8 in high-dose of TCM group and TCM+methotrexate group were increased (P〈0.01, P〈0.05). Conclusion: The resolving stasis and abdominal mass embryo-killing Chinese medicine compound can promote the apoptosis by inducing the Caspase mediated death receptors and mitochondrial apoptosis pathway.
