摘 要:目的:观察青、老年大鼠脑组织miRNA表达谱差异,及补肾方(左归丸)和益气方(益气聪明汤)的调整作用。方法:以自然衰老大鼠(24月龄)为衰老模型,随机分为老年对照组、左归丸组、益气聪明汤组;另设青年对照组(5月龄)。用芯片技术检测各组大鼠脑组织miRNA差异表达变化,并进行实时定量PCR验证,以及利用生物信息软件进行靶基因预测分析。结果:共检测出761种miRNA,与青年对照组比较,老年对照组差异表达的miRNA有126种。左归丸和益气聪明汤干预老年大鼠差异表达的miRNA分别有miR-152-3p、miR-450a-5p等8种和miR-19a-3p、miR-203a-3p等21种。PCR验证结果与芯片数据一致。生物信息软件分析显示左归丸和益气聪明汤干预衰老大鼠差异表达miRNA的靶基因,涉及调控脑功能活动的多条信号通路。结论:青、老年大鼠脑组织miRNA表达有显著差异,补肾益气方药对老年大鼠差异表达的部分miRNA具有改善作用,推测这可能是补肾益气方药延缓脑功能退化的部分作用机制。Objective: To observe the difference of miRNA expression in the brain tissue between young and old rats, and the effects of reinforcing kidney formula (Zuogui Pills) and nourishing qi formula (Yiqi Congming Decoction) on aging rats. Methods: Naturally aging rats (24 month old) were randomly divided into three groups: the old control group, the Zuogui Pills group, and the Yiqi Congming Decoction group. There was a young control group (5 month old) in addition. The expression of miRNA in the brain tissue of each group was detected by the chip technique. Real-time qPCR was used to further validate the expression levels of miRNA. The bioinformatics tools were used to cluster the chip results and predicted the target genes. Results: A total of 761 kinds of miRNA were detected. Compared with the young control group, 126 kinds of miRNA were differentially expressed in the old control group. During the process of aging intervention, there were 8 kinds (miR-152-3p, miR-450a-5p, etc.) and 21 kinds (miR-19a-3p, miR-203a-3p, etc.) of miRNA were differentially expressed in Zuogui Pills group and Yiqi Congming Decoction group respectively. The results of PCR were in accordance with the miRNA array results. Bioinformatics analysis showed that multiple signaling pathways were involved in the in the process of miRNA-targeting regulation. Conclusion: Zuogui Pills and Yiqi Congming Decoction could improve the differentially expressed miRNA in aging rats. It was presumed that this might be part of the mechanism of reinforcing kidney and nourishing qi formula to retard the degeneration of brain function.
