摘  要:目的:研究安神定志灵对SHR大鼠多动冲动行为及前额叶皮质单胺类神经递质及其代谢产物的影响。方法:依据随机数字表将SHR大鼠分为模型组,择思达组(0.045mg/kg),安神定志灵低、中、高剂量组(6.7,13.4,26.7g/kg),每组8只,另设WKY大鼠、SD大鼠各8只为正常组,灌胃给药28d。旷场实验观察大鼠运动距离及运动速度,微透析联合HPLC-ECD检测前额叶去甲肾上腺素(NE)、多巴胺(DA)和代谢产物3,4二羟基苯乙酸(DOPAC)、高香草酸(HVA)含量。结果:(1)给药前,模型组、择思达组及安神定志灵各剂量组运动距离及运动速度较正常组1(WKY)显著升高(P﹤0.05);给药第14、21、28天,择思达组和安神定志灵中剂量组上述指标较模型组显著降低(P﹤0.05)。(2)给药前,模型组、择思达组及安神定志灵各剂量组NE、DA、DOPAC、HVA含量较WKY组显著降低(P﹤0.05);模型组NE、HVA含量较正常组2(SD)显著降低(P﹤0.05);给药第14天,择思达组NE、DA含量较模型组显著升高(P﹤0.05);给药第28天,择思达组、安神定志灵中剂量组NE、DA含量较模型组显著升高(P﹤0.05),安神定志灵中剂量组DOPAC含量较模型组显著升高(P﹤0.05)。结论:安神定志灵能调控SHR多动、冲动行为,改善NE、DA缺乏状态。Objective: To study the effects of Anshen Dingzhi Ling (ADL) on hyperactive and impulsive behavior and levels of monoamine neurotransmitters and metabolites in prefrontal cortex (PFC) of SHR. Methods: SHR rats were randomly divided into model group, atomoxetine (ATX) group (0.045mg/kg), and ADL low, middle, and high dose groups (6.7, 13.4, 26.7g/ kg respectively), 8 in each group. Eight Wistar Kyoto (WKY) rats and eight SD rats were used as normal groups. Rats in various groups received corresponding drugs by ig administration for 28 days. The open field test was used to observe moving distance and speed of the rats. Microdialysis combined with HPLC-ECD was used to detect the contents of NE, DA, DOPAC and HVA. Results: (~)The model group, ATX group and ADL groups had significantly longer moving distance and higher moving speed than the normal group 1 (WKY) before administration (P〈0.05). Compared with the model group, the ATX group and the ADL middle dose group had significant reductions in moving distance and moving speed at 14th, 21st and 28th day after administration (P〈0.05). (~)Before administration, the levels of NE, DA, DOPAC and HVA in the model group and ATX group were significantly lower compared with those in the WKY group (P〈0.05), and the levels of NE and HVA were significantly lower than those in the normal group 2 (P〈0.05). The levels of NE and DA in the ATX group after 2 weeks of treatment were significantly higher than those in the model group (P〈0.05). After 4 weeks of treatment, the levels of NE and DA in the ATX group and ADL middle dose group were significantly higher than those in the model group (P〈0.05), and the level of DOPAC in the ADL middle dose group was significantly higher than those in the model group (P〈0.05). Conclusion: ADL can regulate hyperactivity and impulsive behavior of SHR rats and improve NE and DA deficiency.
