摘 要:目的:建立分析不同产地银杏叶多糖中单糖组分变化的方法。方法:色谱柱为Kromasil C18 column(200mm×4.6mm,5μm),乙腈-水(80∶20)为流动相,流速1.0mL/min,柱温:30℃,进样量:10μL。ELSD漂移管温度80℃,雾化气体压力40psi,喷雾器温度36℃,载气流速1.30mL/min,主成分分析法(PCA)对其单糖进行分析和评价。结果:银杏叶多糖由鼠李糖、果糖、阿拉伯糖、甘露醇、葡萄糖以及半乳糖组成;不同产地银杏叶多糖水解以后单糖总量含量差异较大,单糖总量最高为山东泰安产银杏叶(32.724mg/g),最低为四川南台产银杏叶(20.907mg/g),6个单糖中葡萄糖和阿拉伯糖含量偏高,分别为8.37和8.05mg/g,果糖含量最低(0.746mg/g)。基于糖类成分考虑,银杏叶的分布没有明显的地域性。结论:本研究建立了测定银杏叶多糖组成成分的分析方法,该方法操作简单,方便可行。Objective: To establish an HPLC-ELSD method for determination of polysaccharides composition and the monosaccharide content from Ginkgo biloba leaves. Methods: The analysis was performed on Kromasil C18 column (200mm×4.6mm, 5 μ m) chromatographic column, and the mobile phase was acetonitrile-water (80 : 20). The flow rate of the mobile phase was 1.0mL/min, and the column temperature was maintained at 30℃. The analytes were monitored with ELSD. The drift tube temperature of the ELSD was set at 80℃, the atomization gas pressure was 40psi, sprayer temperature was 36℃, the flow of carrier gas was 1.30mL/min. Furthermore, the principal component analysis (PCA) was applied to the different G. biloba leaves samples. Results: The polysaccharide in Ginkgo biloba leaves consists of rhamnose, fructose, arabinose, mannitol, glucose and galactose. The content of total sugar in the leaves from different areas were different. The total monosaccharides in Shandong Taian was highest (32.724mg/g), while in Sichuan Tainan was lowest (20.907mg/g). The contents of glucose and arabinose (8.37 and 8.05mg/g, respectively) were higher than others. The fructose was lowest (0.746mg/g). Furthermore, the results of PCA implied that there was no correlation between contents and cultivation regions. Conclusion: This method is simple and rapid, stability and feasible, it can be used for the quality control of polysaccharide compositions in Ginkgo biloba leaves.
