摘 要:目的:通过醋制南五味子不同化学部位对2型糖尿病(T2DM)大鼠糖脂代谢的影响研究,筛选醋制南五味子调节T2DM大鼠糖脂代谢的活性部位。方法:采用高脂饲料加链尿佐菌素联合诱导的T2DM大鼠模型,通过测定血清中血糖(FBG)、糖耐量(OGTT)、血清胰岛素(FINS)、游离脂肪酸(FFA)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、谷丙转氨酶(ALT)、谷草转氨酶(AST)的变化、测定肝脏和肾脏组织中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)的变化,观察肝脏、胰腺和肾脏组织病理切片分析组织形态学变化,从而评价醋制南五味子石油醚、氯仿、正丁醇和剩余4个化学部位调节T2DM大鼠糖脂代谢药效学差异。结果:醋制南五味子石油醚、氯仿、正丁醇和剩余部位各剂量组和吡格列酮组均能不同程度地改善T2DM大鼠糖脂代谢。与模型组比较,石油醚、氯仿和吡格列酮组HDL-C、SOD、CAT、GSH-Px显著升高,FFA、LDL-C、TC、TG、ALT、AST、MDA显著降低,但对FBG、OGTT、FINS改善作用不大。结论:醋制南五味子石油醚和氯仿部位对T2DM大鼠肝损伤和高脂状态有显著的改善作用。Objective: To study the effects of different chemical parts of South Schisandra with vinegar on glucose and lipid metabolism in rats with type 2 diabetes mellitus (T2DM) induced by high fat diet and streptozotocin (STZ), and to screen active sites of South Schisandra with vinegar on glycolipid metabolism in T2DM rats. Methods: A rat model of T2DM was induced by high fat diet plus STZ. The changes of FBG, OGTT, FINS, FFA, TC, TG, HDL-C, LDL-C, ALT and AST in serum were measured. The GSH-Px, SOD, MDA and CAT were measured in liver and kidney tissues, and the histopathological changes of liver, pancreas and kidney were observed. Based on these results, the pharmacodynamics differences of petroleum ether, chloroform, n-butanol and four remaining chemical sites on glucose and lipid metabolism in T2DM rats were evaluated. Results: The levels of glycol-lipid metabolism in T2DM rats were improved in different doses of petroleum ether group, chloroform group, n-butanol group, raffinate group and pioglitazone group. Compared with model group, HDL-C, SOD, CAT, GSH-Px were significantly increased and FFA, LDL-C, TC, TG, ALT, AST and MDA were decreased in petroleum ether group, chloroform group and pioglitazone group, but FBG, OGTT, FINS were not improved obviously. Conclusion: Ether and chloroform of South Schisandra with vinegar can significantly improve the liver iniury and hyperlipidemia in T2DM rats.
