摘 要:目的:研究穴位埋线对脑缺血再灌注后肢体痉挛大鼠谷氨酰胺合成酶(GS)表达的影响,为穴位埋线在康复临床中治疗中风后肢体痉挛提供理论依据。方法:制作脑缺血再灌注后肢体痉挛模型,Zea1onga神经功能缺损症状评分1-3分,且改良Ashworth肌张力评分1~4级者,根据随机数字表法分别入组模型组和穴位埋线组,假手术组只分离颈部相关血管;造模3d后,穴位埋线组给予穴位埋线治疗,其他两组在同等条件下抓取,不进行治疗;治疗7d后,分别进行TTC染色,BL-420F电生理记录仪测试肌张力,免疫荧光染色法和Wesern blot法检测3组大鼠脑组织中GS表达的情况。结果:TTC染色显示,治疗后穴位埋线组脑梗死体积明显小于模型组同期(P﹤0.05);BL-420F电生理记录仪测试肌张力显示,治疗后穴位埋线组肌张力较模型组显著升高(P﹤0.05);免疫荧光染色法及Wesern blot法均显示,穴位埋线组较模型组脑组织中GS的表达显著增强(P﹤0.05)。结论:穴位埋线改善脑缺血再灌注大鼠肢体痉挛的作用机制,可能与提高大鼠脑组织中GS的表达有关。Objective: To observe the effects of catgut implantation in acupoints (CIA) on GS expressions in brain of rats with spasticity after stroke, for providing theoretical basis for rehabilitation therapy after stroke by catgut implantation in acupoint. Methods: The stroke model was established by middle cerebral artery occlusion (MCAO). After modeling, the rats were selected into catgut implantation in acupoints group (CIAP) and model group (MP), which were both 1-3 scores of Zealonga neurological impairment scale and 1-4 levels of Modified Ashworth Scale (MAS). Sham group(SP) were only dissected some carotid arteries. Three days after modeling, ClAP was managed by CIA, MP and SP were given no therapy, but fixed by the same way. Before and after therapy, BL-420F physiological recorder was used to measure the muscle tone of quadriceps femoris of rats. The cerebral infarction size was assessed by TTC staining, and GS was detected by immunofluorescence staining and Western blot in three different groups. Results: TTC staining showed that the volum of ClAP was smaller than MP after treatment (P〈0.05). The results of BL-420F showed that muscle tone in CIAP was increased obviously compared with MP (P〈0.05). The expression of GS was significantly higher in CIAP compared with MP (P〈0.05). Conclusion: The mechanism of ClAP in improving limb spasm after cerebral ischemia-reperfusion may be related to the increase of GS expression in rat brain.
