摘 要:目的:探究化橘红对特发性肺纤维化(IPF)的作用及机制。方法:将SPF级大鼠随机分为假手术组、模型组、化橘红组和地塞米松组(Dex组),采用博莱霉素气管滴入方法建立肺纤维化模型,造模后第2天分别给予相应剂量药物灌胃,连续灌胃28d,观察各组肺组织形态、微血管密度(MVD)、肺小动脉管壁厚度、成纤维细胞生长因子(bFGF)及血小板第四因子(PF4)的表达。结果:模型组出现典型肺间质纤维化病变,化橘红组及Dex组明显轻于模型组(P﹤0.01),与模型组相比,化橘红组MVD低于模型组(P﹤0.05),且能明显抑制肺小动脉管壁增厚,其bFGF表达显著低于模型组(P﹤0.01),能明显促进PF4表达(P﹤0.01)。结论:化橘红可能通过抑制IPF大鼠bFGF表达和促进PF4表达而抑制血管新生,进而抑制IPF进程。Objective: To explore the effects and mechanism of Citri Grandis on angiogenesis of idiopathic pulmonary fibrosis rats. Methods: Healthy male SPF rats were randomly divided into four groups: normal control group, model group, Citri Grandis group and Dex group. Establishing the idiopathic pulmonary fibrosis models by dropping bleomycin into trachea, from the second day after model established, each drug treatment group was administered with corresponding drugs for 28 days. The pathological form in lung tissue was observed, the levels of microvessel density (MVD), pulmonary arterial wall thickness, basic fibroblast growth factor (bFGF) and platlet factor 4 (PF4) were detected. Results: The model group had typical pulmonary fibrosis, the Citri Grandis group and Dex group were significantly lighter than the model group (P〈0.01). Compared with the model group, the MVD of the Citri Grandis group was lower than that of the model group (P〈0.05), and the pulmonary arterial wall thickening was obviously inhibited, the expression of bFGF was significantly lower than that of the model group (P〈0.01), and it promoted the PF4 expression (P〈0.01). Conclusion: Citri Grandis can inhibit angiogenesis by inhibiting the expression of bFGF in IPF rats and promoting the expression of PF4 to suppressed the IPF process.
