摘 要:目的观察活血化痰方的降脂效果并探索其降脂的分子机制。方法 SD大鼠36只(180~200 g)据体质量和随机数字表完全随机区组分组,分为6组(n=6):空白组、模型组、可定组、活血化痰方低剂量组、活血化痰方等效剂量组、活血化痰方+可定组。空白组予基础饲料,其余各组均予高脂饲料。造模的同时各药物组按人-动物药物剂量换算表分别给予相应药物灌胃。8周后,观测各组大鼠体质量的变化;生化法检测大鼠血清总胆固醇(TC)、三酰甘油(TAG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)的水平;ELISA法检测大鼠血清前蛋白枯草酶溶菌素9(PCSK9)、血浆卵磷脂胆固醇酰基转移酶(LCAT)、胆固醇酯转移蛋白(CETP)、肝脂酶(HL)的水平及HE染色观察肝脏的病理变化。结果 1)体质量:各组别大鼠体质量差异无统计学意义(P>0.05)。2)血脂:与空白组比较,模型组大鼠血清的TC、TAG、LDL-C水平显著上升(P<0.05);与模型组比较,各药物组大鼠血清的TAG、LDL-C水平均下降(P<0.05);与模型组比较,可定组与活血化痰方+可定组的TC显著下降(P<0.05)。3)关键靶酶:与模型组比较,活血化痰方低剂量组、活血化痰方等效剂量组、活血化痰方+可定组的PCSK9水平均下降(P<0.05),活血化痰方等效剂量组的HL有升高的趋势。4)肝脏的病理变化:与模型组比较,各药物组肝小叶和肝细胞索的结构较清晰,肝细胞内脂质沉积和肝脏脂肪变性程度明显减轻;可定组出现明显的肝细胞肿胀现象。结论活血化痰方可有效降低高脂血症SD大鼠血清TAG、LDL-C水平,其机制可能与其调节血清PCSK9水平、升高血清HL水平有关,同时活血化痰方能够显著改善高脂血症大鼠肝脏的脂质沉积。Objective: To investigate the effect and meehanisnt of Huoxue Huatan Decoction on blood lipids metabolism in hyperlipoidenfic rats. Methods: 36 SD rats were randomly divided into 6 groups (n=6),respec- tively,the blank group,the model group,rosuvastatin group,Huoxue Huatan Decoction of low and high dose groups,and rosuvastatin+Huoxue Huatam Decoction group (R+H group). The blank group was ted with basal diet, while the rest groups were fed with high fat diet. At the same time, all the drug groups were given corresponding drugs. After 8 weeks,the changes of weight of rats were respectively observed;the levels of TC,TG,LDL-C and HDL-C were detected with bioehenfieal method;the levels of PCSK9,LCAT,CETP and HL was detected with ELISA,and the pathological changes of the liver were observed by HE staining. Results: 1)Weight:There was no significant difference in groups (P〉 0.05). 2)Blood fipids: Compared with the blank group,the sermn levels of TC, TG and LDL-C in the model group were significantly increased (P〈 0.05). Compared with the model group,the serum TG and LDL-C of the rats decreased in each drug group (P〈 0.05) ;compared with the model group ,TC of rosuvastatin group and R+H group decreased significantly (P〈 0.05). 3)Key target enzyme:Compared with the model group,the PCSK9 of the low-dose group,the high-dose group and R+H group decreased (P〈 0.05). HL of the high-dose group had an increasing trend. 4)Liver pathological aspects: Compared with the model group,the structures of liver lobules and hepatic cords in drug groups were more clear,and lipid deposition in hepatoeytes and liver steatosis degree were significantly improved ;hepatoeytes of rosuvastatin group were swelling. Conclu-sion: Huoxue Huatau Decoction can reduce sermn leves of TAG and LDL-C in hyperfipoidemie rats,which may be closely related to reducing the serum level of PCSK9 and raising the serum level of HL;meanwhile,the liver lipid deposition of hyperfipoidemic rats can
