四逆汤对高脂血症合并动脉粥样硬化兔模型血清炎性因子的影响

摘　　要：目的观察四逆汤对高脂血症(HLP)合并动脉粥样硬化(AS)兔血清相关炎性因子的影响,探讨其抗动脉粥样硬化的作用机制。方法采用3月龄新西兰大耳白兔60只,随机分为空白组、模型组、阿托伐他汀组及四逆汤低、中、高剂量组,每组10只。除正常组外,其余各组通过高脂饲料喂养建立兔HLP合并AS模型。除空白组和模型组外,其余各组予药物干预。于给药4周后取材,ELISA法检测白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)的表达。结果与正常组比较,模型组IL-6、TNF-α、IFN-γ表达升高(P<0.01),IL-10表达降低(P<0.01);与模型组比较,四逆汤高、中剂量组和西药组IL-6、TNF-α、IFN-γ表达降低(P<0.05或P<0.01),IL-10表达升高(P<0.05或P<0.01)。结论四逆汤具有抗炎、保护血管内皮的作用,其中以四逆汤高剂量组效果最明显。Objective： To observe the effect of Sini Decoction on the inflamnlatory- cytokines associated with atherosclerosis （AS） rabbits with hyperlipidemia （HLP） ,and to explore its mechanisnl of anti-atherosclerosis. Methods： 60 New Zealand rabbits were randomly divided into the blank group,the model group,atotwastatin ＋ model group,Sini Decoction groups （the low dose group, the middle dose group and the high dose＋model group）, 10 case in each group. Except the notxnal group,the other groups established the rabbit model with HLP and AS by feeding high fat diet with inmmne injury method. Except the blank group and the model group,the other groups were given drug intervention. After 4 weeks of administration,ELISA was used to detect the expression of IL-6,IL-10,TNF-α and IFN-γ. Results： Compared with the blank group,the expression of IL-6,TNF-α and IFN-γ in the model group increased（P〈 0.01 ）,and the expression of IL-10 decreased（P〈 0.01 ）. Compared with the model group,the expression of IL-6,TNF-α and IFN-γ in the high dose group,the middle dose group and a- torvastatin＋model group decreased （P〈 0.05 or P〈 0.01） ,and the expression of IL-10 increased （P〈 0.05 or P〈 0.01）. Conclusion： Sini Decoction has an anti-inflammatots- and protective effect on vascular endothelium,and the eftect is most obvious in the high-dose group.
